黄瓜CsGA20ox1异源表达促进拟南芥植株发育 被引量:5

摘要 GA20ox1在植物的种子萌发和植株生长发育过程中起着重要的作用,而黄瓜CsGA20ox1基因在植株生长发育的作用尚不明确。本实验以黄瓜和拟南芥为材料,采用同源克隆和PCR技术分离克隆了黄瓜赤霉素20-氧化酶基因CsGA20ox1,对黄瓜CsGA20ox1基因在植株生长发育的作用进行了初步探究。系统进化树分析显示,CsGA20ox1与At GA20ox1亲缘关系最近;基因表达分析表明CsGA20ox1在黄瓜幼叶和开花当天的子房中表达量较高。亚细胞定位表明黄瓜CsGA20ox1同时在细胞核和细胞质中表达。通过农杆菌介导的遗传转化获得CsGA20ox1超量表达拟南芥转基因植株,拟南芥转基因植株的发芽速率得到显著提高,并且莲座叶面积、叶片面积和叶柄长度显著增大。DIC实验统计结果表明CsGA20ox1增大了表皮细胞面积。 CsGA20ox1 plays an important role in the plant seed germination and plant growth and development process.But the role of CsGA20ox1 in cucumber growth and development is not yet clear.Using cucumber and Arabidopsis thaliana as materials,CsGA20ox1 gene of cucumber gibberellin 20-oxidase was isolated and cloned by homologous cloning and PCR technique.The effects of cucumber CsGA20ox1 gene on plant growth and development were preliminarily explored.Phylogenetic tree analysis showed CsGA20ox1 had the highest evolutionary relationship with AtGA20ox1.Gene expression analysis showed CsGA20ox1 expression was higher in cucumber young leaves and in the ovary of the flowering day,the expression of CsGA20ox1 in flower and tendril tissues was lower than other tissues.Subcellular localization indicated cucumber CsGA20ox1 was expressed in both the nucleus and the cytoplasm at the same time.CsGA20ox1 overexpression of A.thaliana transgenic plants were obtained by agrobacterium mediated genetic transformation.The germination rate of A.thaliana transgenic plants was significantly increased.The rosette leaf area,leaf area and petiole length increased significantly.And the rosette area and leaf area increased significantly.DIC statistical results showed CsGA20ox1 increased the area of epidermal cells.
