摘要
昆虫体内栖息着大量的微生物,对宿主生长发育、生殖、营养代谢以及免疫等方面均起着重要作用。目前,昆虫内生菌的鉴定方法有传统的鉴定方法和变性梯度凝胶电泳(DGGE)、16Sr RNA基因技术、分子杂交技术、限制性片段长度多态性(RFLP)、扩增性核糖体DNA限制性酶切片段分析(ARDRA)、实时荧光定量PCR技术(RT-q PCR)以及宏基因组测序技术等分子生物学方法。本文对这些鉴定方法在昆虫研究领域的应用进行概述,为昆虫内生菌的研究提供技术参考。
Many microrganisms were inhabited in insects and played an important role in host growth and development,reproduction,nutritional metabolism and immunity etc.At present,insect symbiotic bacteria were identified using traditional and some molecular biology methods,such as denaturing gradient gel electrophoresis(DGGE),16SrRNA gene technology,molecular hybridization technology,restriction fragment length polymorphism(RFLP)analysis,amplified ribosomal DNA restriction fragments analysis(ARDRA),real-time fluorescence quantitative PCR(RT-qPCR)and macro genome sequencing technology etc.This paper summarizes the application of these identification methods in the field of insect research,and provides a technical reference for the study of insect symbiotic bacterium.
