信号调节蛋白α1在大鼠肝再生过程中表达的实验研究 被引量:2

摘要 目的 观察信号调节蛋白α1 (SIRPα1 )在大鼠肝再生过程中的表达变化。方法 选取雄性SD大鼠 1 2 0只 ,随机分为两组 :假手术组 (SO)和肝切除手术组 (OP) ,每一组又分为 1 0个时间点 ,即手术后 2、6、1 2、2 4、30、48、72、1 2 0、1 68和 2 4 0h ,每一时间点 6只 ,切除大鼠 70 %肝脏 (肝左叶和中叶 )建立肝再生模型 ,术后在预定时间点分别取肝组织固定石蜡包埋切片 ,采用免疫组织化学方法测定肝组织SIRPα1表达。结果 再生肝组织 1 2h肝实质可见局灶散在肝细胞膜棕黄色着色 ,2 4h弥漫性肝细胞膜着色 ,1 2 0h以后肝细胞膜着色逐渐减弱。结论 SIRPα1作为一种负向调控因子参与了肝细胞再生 ,可能与肝再生终止有关 ,但关于其详细调控机制及其与其他因子相互作用的机制有待于进一步研究。 Objective to observe the expression change of signal regulatory protein α1 on rat hepatic regeneration.Methods 120 Sprague Dawley male rats were selected and divided into 2 groups randomly,sham operation group(SO)and hepatectomized group(OP),each group was 10 indicated time points,including 2,6,12,24,30,48,72,120,168,240?h postoperatively,6 rats were in every time point;70% of liver including media and left lobes were resected in hepatectomized group,all animals were sacrificed at the indicated time points and hepatic tissues were fixed and embedded into paraffin and sectioned.The expression of SIRP α1 in hepatic tissue was determined by immunohistochemistry.Results The brown yellow staining of hepatic cellular membrane in hepatic parenchyma was locally observed at 12?h,the diffuse staining of hepatic cellular membrane in hepatic parenchyma was observed at 24?h,and the staining became gradually light after 120 h.Conclusion SIRPα1,as a negative regulatory factor,participates in hepatocytes proliferation,and may be involved in termination of hepatic regeneration,but it is needed to further study on the detailed regulating mechanism and the mechanism of interacting with other regulators.
