噬菌体vB_SalS_DZ的生物学特性及对鸡沙门氏菌病的疗效 被引量:6

摘要 抗药性沙门氏菌的出现,严重影响了抗生素对沙门氏菌病的治疗效果,而噬菌体治疗是一种可能的替代方法。本试验研究了沙门氏菌噬菌体vB_SalS_DZ的基本生物学特性及其对沙门氏菌SM86的治疗效果。噬菌体vB_SalS_DZ的生物学特性分析显示其效价可达109pfu/m L,透射电镜结果显示该噬菌体属于有尾目T4噬菌体。对p H值的适应范围宽,在p H值4~12内能保持较高的裂解活性;该噬菌体的最适p H值在5.0左右。最佳感染复数为10-6,感染宿主菌的潜伏期和爆发期均为60 min;基因组中不携带抗性基因和毒力基因。噬菌体vB_SalS_DZ可以使感染沙门氏菌病的SPF鸡死亡率降低50个百分点,这表明该噬菌体在治疗特定的沙门氏菌感染中具有较大的应用潜能。 The emergence of drug-resistant Salmonella seriously affected the therapeutic effect of antibiotics on salmonellosis,and bacteriophage therapy was a possible alternative.This study explored the basic biological characteristics of Salmonella phage vB_SalS_DZ and its therapeutic effect on Salmonella SM86.The biological characteristics of Salmonella phage vB_SalS_DZ showed that the titer was 10 9 pfu/mL.The results of transmission electron microscopy showed that the phage belonged to the T4 phage.It had a wide adaptation range to pH,and maintained high cleavage activity at pH 4~12.The optimum pH of the phage was about 5.0.Its optimal multiplicity of infection was 10-6,and the incubation period and outburst period were both about 60 min.The genome did not carry resistance genes and virulence genes.Animal experiments showed that phage vB_SalS_DZ could reduce the mortality of SPF chicken infected with salmonellosis by 50 percent points,which indicated that the phage vB_SalS_DZ had great potential in the treatment of specific Salmonella infection.
