血府逐瘀汤对冠心病血瘀模型家兔氧化应激的影响

Effects of Xuefu Zhuyu Decoction on Oxidative Stress in Rabbits with Blood Stasis Syndrome of Coronary Heart Disease

为观察血府逐瘀汤对冠心病(CHD)血瘀模型家兔氧化应激的影响,将清洁级健康家兔随机分为对照组,模型组,假手术组,血府逐瘀汤低剂量、中剂量、高剂量6组,每组8只。模型组、低剂量、中剂量、高剂量组采用左冠状动脉放置缩窄环方法,制备冠心病模型。低剂量、中剂量、高剂量组以血府逐瘀汤按10、20、40g/kg体重灌胃,每天1次,连续21d。对照组、模型组、假手术组则给予等体积生理盐水。检测血管紧张素Ⅱ(AngⅡ)、活性氧(ROS)、肌酸激酶(CK)、乳酸脱氢酶(LDH)、总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、丙二醛(MDA)水平、NADPH氧化酶亚型2(NOX2)表达。结果表明,与对照组比较,模型组AngⅡ、ROS、CK、LDH、MDA水平显著或极显著升高(P<0.05或P<0.01),TAOC、SOD、GSH水平显著或极显著下降(P<0.05或P<0.01),NOX2表达增多。与模型组比较,中剂量、高剂量组AngⅡ、ROS、CK、LDH、MDA水平显著或极显著降低(P<0.05或P<0.01),而T-AOC、SOD、GSH水平显著或极显著升高(P<0.05或P<0.01),NOX2表达减少。提示血府逐瘀汤具有减轻心肌细胞氧化损伤作用,可能是其治疗CHD的部分起效环节。

To investigate the effects of Xuefu Zhuyu decoction on oxidative stress in the rabbits with blood stasis syndrome of coronary heart disease,clean grade healthy rabbits were randomly divided into control group,model group,sham group,Xuefu Zhuyu decoction low-dose,middle-dose,high-dose groups with eight rabbits in each group.Left coronary artery implanted constrictor ring to establish the model of coronary heart disease in the model group,the low-dose,the middle-dose and the high-dose groups.The low-dose,the middle-dose,the high-dose groups were intragastrically administrated with Xuefu Zhuyu decoction according to 10,20,40 g/kg respectively,once a day for continuous 21 days,while the control group,the model group and the sham group with the same volume normal saline,then the levels of AngⅡ,ROS,CK,LDH,T-AOC,SOD,GSH,MDA and the expression of NOX2 were detected.The results showed that compared with the control group,the levels of AngⅡ,ROS,CK,LDH,MDA increased(P<0.05 or P<0.01)while the levels of T-AOC,SOD,GSH reduced(P<0.05 or P<0.01),the expression of NOX2 increased in the model group,and compared with the model group,the levels of AngⅡ,ROS,CK,LDH,MDA reduced(P<0.05 or P<0.01)while the levels of T-AOC,SOD,GSH increased(P<0.05 or P<0.01),the expression of NOX2 reduced in the middle-dose and the high-dose groups.It was supposed that Xuefu Zhuyu decoction could alleviate myocardial cell oxidative injury,which could be a part of mechanisms of treatment of CHD.
