摘要
为了促进环介导等温扩增(LAMP)技术的推广应用,在传统LAMP反应体系中引入羟基萘酚蓝指示剂,建立可视LAMP方法,对反应体系进行高温-低温前处理和添加甜菜碱等优化措施提高检测敏感性,液面覆盖矿物油降低气溶胶污染,据此建立的猴沙门菌可视LAMP检测方法敏感度为6CFU/反应(25μL体系),结果判定直观明了,与其他细菌无非特异性反应,检验效率和检出率均优于细菌培养鉴定法。
In order to promote the application of LAMP technology,the indicator hydroxynaphthol blue was added to reaction system of the traditional LAMP in the study.The reaction system was prepared by heating-cooling pretreatment,and betaine was added to the reaction system to increase the sensibility.The liquid surface was covered by mineral oil to decrease aerosol contamination.Base on those above,we built a visible loop-mediated amplification method for detecting experimental monkey Salmonella.The sensibility of the visible LAMP is 6 CFU/reaction.The visible LAMP showed intuitive and specific results,more efficient and higher detection rate than germiculture.
