氧化苦参碱对病毒性心肌炎小鼠巨噬细胞移动抑制因子的影响研究 被引量:5

摘要 目的:通过观察氧化苦参碱(OMT)对小鼠病毒性心肌炎(VMC)心肌的保护作用,探讨巨噬细胞移动抑制因子(MIF)在病毒性心肌炎发病机制中的作用及氧化苦参碱对其的干预作用。方法:选取60只4周龄Balb/c小鼠随机分为4组,分别为空白组、对照组及高剂量OMT组、低剂量OMT组(n=15),经腹腔CVB3制成病毒性心肌炎模型,氧化苦参碱灌胃观察其对小鼠病毒性心肌炎的作用。小鼠心肌切片苏木精-伊红染色、观察心肌坏死和炎症细胞浸润面积的变化。RT-PCR检测小鼠心脏MIF mRNA表达。结果:MIF mRNA表达水平在对照组表达量高于空白组(P<0.05);在氧化苦参碱干预后MIF mRNA表达水平,高剂量OMT组低于低剂量OMT组(P<0.05);高剂量OMT组、低剂量OMT组小鼠心肌坏死组织面积与总面积比值低于对照组(P<0.05)。高剂量OMT组心肌炎症细胞浸润与总面积比值低于低剂量OMT组(P<0.05)。结论:OMT可改善VMC小鼠的心肌炎症,下调MIF在心肌的表达,表明OMT对VMC心肌产生保护作用可能与下调MIF的表达有关。 Objective:By observing the protective effect of oxymatrine(OMT)on myocarditis in mice with viral myocarditis(VM),the role of macrophage migration inhibitory factor(MIF)in the pathogenesis of viral myocarditis and therapeutic effect of oxymatrine were investigated.Methods:Sixty 4-week-old Balb/c mice were randomly divided into 4 groups:blank group,control group,high OMT dose group and low OMT dose group(n=15),viral myocarditis model was made by intraperitoneal coxasckievirus B3(CVB3),observation of the effect of oxymatrine on mouse with VM by gavage.Hematoxylin-eosin staining of mouse myocardial slice was used to observe the changes of ventricular myocyte necrosis and infiltration area of inflammatory cells.The expression levels of myocardial MIF mRNA were detected by RT-PCR.Results:The expression level of MIF mRNA in the control group was higher than that in the blank group(P<0.05),for the MIF mRNA expression level after oxymatrine intervention,the high-dose OMT group was lower than the low-dose OMT group(P<0.05),the ratio of myocardial necrosis tissue area to total area in high-dose OMT group and low-dose OMT group was lower than that in control group(P<0.05),the ratio of infiltration of inflammatory cells to total area of myocardium in high-dose OMT group was lower than that of low-dose OMT group(P<0.05).Conclusion:OMT can improve the myocardial inflammation and survival rate in VMC mice,and down-regulate the expression of MIF in the myocardium,indicating that the protective effect of OMT on VMC myocardium may be related to the down-regulation of MIF expression.
