环状RNA circ0023033及其靶基因在肺癌中的表达及诊断标志物意义

The aberrant expression of circular RNA circ0023033 and its target genes in lung cancer and its diagnostic significance

目的不同肺癌细胞株中检测circ0023033及其靶向mi RNA、mRNA的表达改变,结合靶基因作为诊断标志物的意义分析,探索新的标志物。方法采用英文文献检索结合GEO及circ Base数据库分析,选择呼吸系统肿瘤新的环状RNA标志物circ0023033,通过RT-qPCR检测其在正常细胞及不同肺癌细胞株中的表达。通过Reg RNA及Targetscan网站预测分析circ0023033靶向的miRNA及mRNA,结合肿瘤标志物的分析,选择差异明显的基因运用RT-qPCR进行检测。结果与BEAS-2B细胞相比,circ0023033在不同肺癌细胞株中明显上调。结合生物信息学分析及RT-qPCR,发现circ0023033可靶向多个mi RNA及mRNA,其中多个为肿瘤诊断标志物,且在肺癌中的表达具有显著差异。结论 circ0023033在肺癌中显著高表达,其多个靶基因可作为肿瘤诊断标志物,显示circ0023033具备作为新的肺癌诊断标志物的意义。

Objective To explore new markers by investigating the changes of circ0023033 and its targeted miRNAs and mRNAs in different lung cancer cell lines,and analyze the significance of target genes as diagnostic makers.Methods An English literature review was conducted and combing with analysis of GEO and circBase databases,a new circular RNA marker circ0023033 was selected for respiratory cancer.RT-qPCR was used to detect its expression in normal cells and different lung cancer cell lines.The circ0023033-targeted miRNAs and mRNAs were predicted from the RegRNA and Target scan website.In combination with the analysis of tumor markers,genes with significant differences were selected for RT-qPCR detection.Results Compared with BEAS-2B,circ0023033 was significantly up-regulated in different lung cancer cell lines.Combined with bioinformatics analysis and RT-qPCR discovery,circ0023033 could target multiple miRNAs and mRNAs,and many of them were tumor diagnostic markers which expression in lung cancer was significantly different.Conclusion Circ0023033 is highly expressed in lung cancer.The tumor marker of its target gene indicates the significance of circ0023033 as a new lung cancer diagnosis marker.
