摘要
目的探讨结肠癌患者应用直接测序法测定K-ras基因的灵敏度及其临床价值。方法选取我院2016年2月~2017年4月收治的结肠癌患者80例,随机分为研究组和对照组两组,研究组患者通过直接测序法、对照组患者通过sanger测序法检测患者体内的K-ras基因突变情况,分析K-ras基因突变率与患者肿瘤大小、肿瘤部位、患者年龄等一般资料之间的相关性。结果研究组患者测定K-ras基因突变的灵敏度明显高于对照组(P<0.05),肿瘤直径在5 cm以上患者的K-ras基因突变率明显低于直径在5 cm以下的患者(P<0.05),研究组检出率与患者肿瘤部位及分化程度、患者年龄等存在相关性。结论应用直接测序法检测结肠癌患者K-ras基因的突变情况具有较高的灵敏度,直接测序法的检出率与患者的肿瘤大小、肿瘤分化程度以及患者的年龄等存在较强的相关性,对于临床上结肠癌等恶性肿瘤疾病的诊断具有较强的应用价值,值得临床上广泛推荐使用。
Objective To analyze the sensitivity and clinical value of direct sequencing in the detection of K-ras gene in colon cancer patients.Methods 80 colon cancer patients from February 2016 to April 2017 in our hospital were randomLy divided into the study group and the control group.The K-ras gene mutations in the study group were detected by direct sequencing and those in the control group were detected by sanger sequencing.The correlation of the K-ras gene mutation rate with the patient's general data including tumor size,tumor location and age was analyzed.Results The sensitivity of the K-ras gene mutation detection in the study group was significantly higher than that in the control group(P<0.05).The K-ras gene mutation rate was significantly lower in patients with tumor diameters above 5 cm than that in patients with diameters below 5 cm(P<0.05).The detection rate of the study group is related to the location and differentiation of the patient's tumor and the age of the patient.Conclusion The direct sequencing method in the detection of the mutation of K-ras gene in patients with colon cancer has high sensitivity.The detection rate of direct sequencing is strongly correlated with the patient's tumor size,tumor differentiation,and the patient's age.The method has strong application value for the clinical diagnosis of malignant diseases such as colon cancer,and it is worth recommending.
