基因芯片筛查外周血中胃癌转移相关基因的研究

摘要 目的:探讨基因芯片筛查外周血中胃癌转移相关基因的效果。方法:收集2016年5月-2018年3月,于我院普外科住院确诊为胃癌但是未经手术治疗的30例患者,同时选取15名健康人员,每位患者分别于术前当日、术中、术后第1天3个时间段的清晨采集空腹静脉血,术中选取癌组织、癌旁组织(距肿瘤边缘约为5cm)、正常组织(距肿瘤边缘约为10cm)标本。全部病例均经术后病理结果证实。采用phalanx人全基因组表达谱芯片对胃癌患者空腹静脉血样、胃癌组织、癌旁组织正常组织进行检测,观察容易检测、特异性高的基因。结果:在参与检测的942个基因中,共筛选出69个显著差异表达的阳性基因,其中28个基因表达下调,41个基因表达上调。结论:运用基因芯片能够有效筛查外周血中胃癌转移相关基因,具有显著效果与较高的临床推广价值。 Objective:To explore the effect of gene chip screening for gastric cancer metastasis in peripheral blood.Methods:30 patients diagnosed as gastric cancer but were not treated with surgery in our hospital from May 2016 to March 2018 were selected.15 healthy people were selected.Each patient collected venous blood in the early morning of the preoperative day,during the operation and the first day after the operation.During operation,the tumor tissues,adjacent tissues(about 5cm from the edge of the tumor)and normal tissue(about 10cm from the edge of the tumor)were selected.All cases were confirmed by postoperative pathology.The phalanx whole-genome expression profile was used to detect the fasting venous blood samples,gastric cancer tissues and adjacent tissues of gastric cancer patients.The genes that were easy to detect and have high specificity were observed.Results:Of the 942 genes involved in the test,69 positive genes with significant differences were selected,28 of which were down-regulated and 41 were up-regulated.Conclusion:The gene chip could effectively screen the related genes of gastric cancer in peripheral blood,and it had significant effects and high clinical value.
