摘要
研究鮸鱼鱼鳔多糖(Polysaccharide of swim bladder in Nibe Croaker,PSB)的单糖组成、分子量、理化性质及其对肝损伤的保护作用。以四氯化碳(CCl4)诱导急性肝损伤小鼠为模型对鱼鳔多糖进行保护肝脏方面研究,测定血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)和谷胱甘肽过氧化物酶(GSH-PX)的活力,并计算肝脏指数并测定肝脏中丙二醛(MDA)、总超氧化物歧化酶(SOD)、总一氧化氮合成酶(NOS)的含量,观察肝脏组织的病理切片。结果表明PSB为富含氨基糖和糖醛酸的硫酸化杂多糖,甘露糖、氨基葡萄糖、葡萄糖醛酸、氨基半乳糖、葡萄糖和半乳糖组成,分子量为89.7kDa。抗肝损伤的实验中与模型组相比,PSB的高剂量组显著地抑制ALT、AST和MDA的升高(P<0.01);PSB高剂量和中剂量组显著地提高GSH-PX活力,SOD和NOS的含量(P<0.01)。综上可见鮸鱼鱼鳔硫酸杂多糖PSB对四氯化碳诱导的小鼠急性肝损伤有一定的保护作用,这一作用可能与提高小鼠自身免疫有关。
To explore the physicochemical property and hepatoprotective effect of the polysaccharides of swim bladder in Nibe Croaker(PSB)on carbon tetrachloride(CCl 4)-induced acute liver injury in mice.The data showed PSB was a sulfated heteropolysaccharide mainly composed of galactosamine glucuronic acid and mannose with the molecular weight of 89.7 kDa.To study the hepatoprotective effect,the alanine aminotransferase(ALT),aspartate aminotransferase(AST)and glutathion peroxidase(GSH-PX)activities in the serum of the mice were determined.The liver index was calculated.The malondialdehyde(MDA),superoxide dismutase(SOD)and nitric oxide(NOS)contents in liver were also studied.Compared with the model group,feeding with high dose group of PSB revealed a significant decrease in the activity of ALT,AST and MDA(P<0.01).The high dose and middle dose group of PSB revealed a significant increase the content in GSH-PX,SOD and NOS(P<0.01).It showed that the sulfated heteropolysaccharides extracted from Swim Bladder in Nibe Croaker could protect CCl 4-induced liver injury in rats,which may be related to enhance the ability of anti-oxidant activities of mice.
