痛泻安脾汤对腹泻型肠易激综合征模型大鼠5-HT和NPY蛋白表达的影响 被引量:6

Effect of Tongxie Anpi decoction on the expression of 5-HT and NPY protein in rats with diarrhea induced irritable bowel syndrome

摘要 目的探讨痛泻安脾汤对腹泻型肠易激综合征模型大鼠5-HT和NPY蛋白表达的影响。方法将40只SD大鼠随机分为空白组、模型组、痛泻安脾汤组和盐酸洛哌丁胺组,各组10只,建立腹泻刑场以及综合征(D-IBS)大鼠模型,采用免疫组化法分别检测大鼠肠组织中NPY和5-HT的蛋白表达水平和ELISA法测定血清NPY和5-HT含量。结果模型组大鼠血清5-HT和NPY含量均较空白组升高,差异有统计学意义(P<0.05);血清5-HT和NPY在各组大鼠水平为模型组>盐酸洛哌丁胺组>痛泻安脾汤组,差异有统计学意义(P<0.05);模型组大鼠回盲部和结肠远端5-HT阳性细胞数量显著较空白组增多,免疫组化染色强度显著增强,差异有统计学意义(P<0.05);痛泻安脾汤组和盐酸洛哌丁胺组大鼠回盲部和结肠远端5-HT阳性细胞数量均较模型组减少,免疫组化染色强度明显降低,差异有统计学意义(P<0.05);大鼠下丘脑、结肠组织NPY蛋白表达水平为空白组>痛泻安脾汤组>盐酸洛哌丁胺组>模型组,差异有统计学意义(P<0.05)。结论痛泻安脾汤可能通过降低腹泻型肠易激综合征模型大鼠血清5-HT、NPY水平,降低肠道5-HT表达,上调模型大鼠下丘脑和肠道NPY蛋白表达而改善腹泻症状。 Objective To investigate the effect of Tongxie Anpi decoction on the expression of 5-HT and NPY protein in rats with diarrhea induced irritable bowel syndrome.Methods Forty SD rats were randomly divided into blank group,model group,Tongxie Anpi decoction group and loperamide hydrochloride group,10 rats in each group,the establishment of diarrhea syndrome and death(D-IBS)rat model by immunohistochemistry was used to detect NPY and 5-HT in the intestinal tissue of rats the protein expression level and determination of serum NPY and 5-HT content by ELISA.Results The serum levels of 5-HT and NPY in the model group were higher than those in the blank group,and the difference was statistically significant(P<0.05).The level of serum 5-HT and NPY in the rats of each group was the model group>the group of droperidine hydrochloride>Tongxie Anpi decoction group,the difference was statistically significant(P<0.05).In the model group,the number of 5-HT positive cells in the ileocecal and distal colon was significantly higher than those in the blank group,and the immunohistochemical staining intensity was significantly increased,the difference was statistically significant(P<0.05).The number of 5-HT positive cells in ileum and distal colon of rats in the Tongxie Anpi decoction Group and the perchloric acid hydrochloride group were lower than those in the model group,and the immunohistochemical staining intensity was significantly decreased,the difference was statistically significant(P<0.05).The expression level of NPY protein in hypothalamus and colonic tissue of rats was blank group>Tongxie Anpi decoction group>the hydrochloride droperidol group>the model group,the difference was statistically significant(P<0.05).Conclusion Tongxie Anpi decoction may improve the symptoms of diarrhea by decreasing the level of serum 5-HT and NPY,reducing the expression of 5-HT in intestinal tract,and up regulating the expression of NPY protein in hypothalamus and intestine of rats with diarrhea predominant irritable bowel syndrome.
