不同年龄段感音神经性聋患者谷氨酸受体7基因多态性分析

Association between GRM7 Gene Polymorphism and Sensorineural Hearing Loss in Different Ages

目的研究不同年龄段感音神经性聋患者代谢型谷氨酸受体7(GRM7)基因rs11928865、rs11920109位点单核苷酸多态性(SNP)及其与年龄的相关性。方法研究对象为906例感音神经性聋患者和755例健康体检者,按年龄段分为试验组(感音神经性聋A组和B组)和对照组(健康体检者C组和D组),A组467例(30~50岁),平均听阈50.91±19.34dB HL,B组439例(51~70岁),平均听阈53.53±18.44dB HL,C组278例(30~50岁),平均听阈9.22±6.39dB HL,D组477例(51~70岁),平均听阈11.33±6.25dB HL。各组分别采集外周血抽提DNA,应用Sequenom MassARRAY方法进行SNP位点分析。对试验组与对照组间的GRM7基因rs11928865、rs11920109位点基因型及等位基因频率进行统计学分析。结果51~70岁年龄段感音神经性聋组与对照组间rs11928865位点基因型分布差异有统计学意义(P=0.018),30~50岁年龄段感音神经性聋组与对照组间差异无统计学意义(P>0.05);rs11920109位点基因型在30~50岁及51~70岁年龄段感音神经性聋组与对照组间差异均无统计学意义(P>0.05)。结论 GRM7基因rs11928865位点多态性与51~70岁年龄段感音神经性聋易感性相关,但与30~50岁年龄段感音神经性聋易感性无相关性。

Objective To study the potential correlations between the single nucleotide polymorphisms(SNPs)rs11928865 and rs11920109 of the gene encoding glutamate metabotropic receptor 7(GRM7)and sensorineural hearing loss in different ages.Methods A total of 1 661 subjects were studied,including patients aged 30~50 and 51~70 years with sensorineural hearing loss and those without hearing loss.Individuals of the 30~50-year-old with sensorineural hearing loss were brought into Group A,those of 51~70-year-old with sensorineural hearing loss as Group B,while the control groups were Group C(30~50-year-old without hearing loss),and group D(51~70-year-old without hearing loss).The Mass ARRAY method was used to analyze genotypes.Results The differences in genotypes for the GRM7 rs11928865 SNP between patients in the 51~70-year-old sensorineural hearing loss group and the control group were statistically significant(P=0.018);the distribution frequencies of genotypes in patients of 30~50 years old were not significantly different(P>0.05);the difference in genotypes for the rs11920109 SNP between the sensorineural hearing loss and control groups showed no statistical significance(P>0.05).Conclusion The rs11928865 SNP was associated with the susceptibility to hearing loss in patients of 51~70-year-old sensorineural hearing loss group,but not with those in the 30~50-year-old sensorineural hearing loss group.
