WIF1在宫颈癌中的表达及其对宫颈癌细胞生物学行为的影响 被引量:6

摘要 目的探讨Wnt抑制因子(WIF1)在宫颈癌中的表达及其对宫颈癌细胞生物学行为的影响。方法采用Western blot及RT-PCR法检测该院经手术切除的82例宫颈癌组织及癌旁组织中WIF1蛋白及mRNA表达,并应用LipofectamineTM2000将pcDNA 3.1 WIF1真核表达载体(过表达组)及空质粒pcDNA3.1(对照组)转染到宫颈癌Hela细胞中,MTT法检测细胞活力,流式细胞术检测细胞凋亡及细胞周期,Transwell法检测细胞侵袭能力,Western blot检测CyclinD1、C-myc、基质金属蛋白酶(MMP)-2及MMP-9表达。结果与癌旁组织比较,宫颈癌组织中WIF1蛋白及mRNA表达量显著降低(P<0.01);与对照组比较,过表达组中WIF1蛋白及mRNA表达量提高(P<0.01),细胞活力降低(P<0.01),细胞凋亡率提高(P<0.01),细胞周期阻滞于G1期(P<0.01),CyclinD1、C-myc、MMP-2、MMP-9表达量显著下调(P<0.01)。结论 WIF1在宫颈癌组织中低表达,提高WIF1表达能显著抑制宫颈癌Hela细胞增殖、侵袭。
