摘要
为了解培养基前后变化和接种不同生长期细菌对印度芽孢杆菌Bacillus indicus L15菌株生长的影响,测定了经2216E(Z)、海水发酵(S)、淡水发酵(F)培养基培养的L15菌株营养细胞和芽孢的生长情况。结果表明:接种由同种培养基培养的L15菌株营养细胞和芽孢后的细菌增长量,并非总是接种营养细胞的增长量高于接种芽孢的增长量,细菌增长量与菌株前期培养用培养基及后期转接培养基有关;接种由淡水发酵培养基培养的L15菌株营养细胞后的细菌增长量显著低于接种芽孢后的增长量(P<0.05),而接种由海水发酵培养基培养的L15菌株营养细胞后的细菌增长量却显著高于接种芽孢后的增长量(P<0.05);当菌株从能被直接吸收的小分子物质组成的培养基转移到不能被直接吸收的大分子物质组成的培养基后,L15菌株增长量显著降低(P<0.05),反之增长量显著升高(P<0.05)。研究表明,在养殖池塘中使用益生菌时需做预试验,以便确定使用营养细胞还是芽孢,且培养益生菌的培养基的碳、氮源组成与使用目标环境和营养分子的组成越接近越好。
To understand the effect of inoculation vegetative cell and spores which were cultured by different media on the growth of Bacillus indicus L15 strain,the growth of L15 strains cultured in the 2216E(Z),seawater(S)and fresh water(F)fermentation media by inoculation spore and vegetative cell were determined.The results showed that the bacterial increase amount were not always higher by inoculation vegetative cell than spore,which were related to medium of early cultivation L15 strain and following medium.The growth rates of L15 strain in above three media were lower by inoculation vegetative cell than spore which were cultured by freshwater fermentation medium,but they were higher by inoculation vegetative cell than spore which were cultured by seawater fermentation medium.The growth rate of L15 strain was decreased when it was transferred to a medium composed of macromolecule organics from a medium composed of micromolecule organics,but it was improved when it was transferred to a medium composed of micromolecule organics from a medium composed of macromolecule organics.In conclusion,the preliminary experiment was required to determine if vegetative cell or spore was used before probiotics were used in the pond.The more similar organic molecules in the medium culturing probiotics in the environment using probiotics were,the best growth the probiotics had.
