摘要
为评估包虫病基因工程亚单位疫苗的安全性和免疫效果,选取承德市接坝地区4个牧场479只8周龄羔羊进行免疫试验。免疫组(308只)于首次免疫前、加强免疫前,以及首次免疫后2、6和9个月,对照组(171只)于试验前和试验后9个月,采集羔羊血清,采用ELISA抗体检测试剂盒进行抗体检测。结果显示:免疫疫苗后,个别羊有轻微的免疫反应;免疫组免疫前抗体阳性率为2.27%,加强免疫前抗体阳性率为87.16%,首次免疫后2、6和9个月的抗体阳性率分别为99.32%、93.49%和86.46%;对照组试验前抗体阳性率为2.34%,9个月后抗体阳性率为7.93%。结果表明,包虫病基因工程亚单位疫苗能够快速诱导机体产生抗体,并且抗体维持期较长,9个月后,动物个体仍维持较高的抗体水平。因此,该亚单位疫苗免疫效果及安全性较好,可以在包虫病流行区推广应用。
In order to evaluate the security and immune effects of the genetically engineering subunit vaccine of hydatid disease,479 8-week-old lambs from 4 pastures in the Bashang area of Chengde City were selected and divided into 2 groups:immunization group(308 lambs)and control group(171 lambs).Then immunological experiments were carried out.For the lambs in the immunization group,serum samples were selected before the first and booster immunization and at the time of 2,6 and 9 months after the first immunization.For the control group,serum samples were collected before the experiment and at the time of 9 months after the experiment.All these serum samples were used for antibody detection by ELISA kits.Results showed that,after vaccination,individual lambs had a slight immune response.For the lambs in the immunization group,the positive rates of the antibody before the first immunization and booster immunization were 2.27%and 87.16%,respectively;besides,at 2,6 and 9 months after the first immunization,the rates were 99.32%,93.49%,and 86.46%,respectively.For the control group,the antibody positive rates before and after the experiment were 2.34%and 7.93%,respectively.Results showed that,the genetically engineering subunit vaccine of hydatid disease could rapidly induce the lambs to produce antibodies,and the antibodies could last for a long time.After 9 months of immunization,the lambs still maintained high antibody levels.Therefore,the echinococcosis vaccine was safe and had good effects,and could be promoted and applied in the endemic area of echinococcosis.
