微囊化嗅鞘细胞移植周围神经损伤痛及下调背根神经节P2X3嘌呤受体的表达

摘要 目的:探讨微囊化与非微囊化嗅鞘细胞移植对周围神经损伤的作用及对P2X3嘌呤受体的表达水平影响。方法:取SD大鼠嗅球组织运用差速贴壁方法并扩增培养嗅鞘细胞。将SD大鼠分成4组,对照组(sham)、慢性坐骨神经损伤模型组(CCI)、非微囊化嗅鞘细胞移植组(non-MC-OECs)及微囊化嗅鞘细胞移植组(MC-OECs);于术后10d和20d运用行为学方法测各组大鼠机械缩足反射阈值;原位杂交组织化学方法检测背根节P2X3受体的阳性细胞百分比及平均吸光度。结果:术后10d和20d,CCI组、non-MC-OECs组、MC-OECs组及sham组大鼠机械缩足反射阈值从低到高;原位杂交检测背根节P2X3受体阳性细胞百分比及平均吸光度依次减低。non-MC-OECs组与CCI组对比,大鼠机械缩足反射阈值明显增高,P2X3受体阳性细胞百分比及平均吸光度明显减低;与non-MC-OECs组比较,MC-OECs组大鼠机械缩足反射阈值增高,P2X3受体阳性细胞百分比及平均吸光度降低,各组间比较差异均有统计学意义。结论:与非微囊化嗅鞘细胞相比,微囊化嗅鞘细胞移植能更好治疗慢性周围神经损伤,减低其背根节P2X3受体表达水平。 Objective:To investigate the effect of microencapsulated and non-microencapsulated olfactory ensheathing cells(OECs)transplantation on peripheral nerve injury and the expression of P2X3 purine receptor.Methods:NASH method was used to cultivate and expand cells from olfactory bulb tissue of a SD rat.SD rats were randomly divided into sham group,model group,non-microencapsulater-OECs(non-MC-OECs group)and microencapsulated OECs group(MC-OECs groups).The 10th and 20th days after surgery,the mechanical stimulation constraining thresholds of the rats in each group were measured by behavioral methods.P2X3 receptor positive cells percentage and average absorbance were observed by in situ hybridization histochemistry.Results:The 10 th and 20 th days after surgery,the mechanical stimulation constraining thresholds of rats were increased from the model group,the non-MC-OECs group,the MC-OECs group to the sham group,and the positive cells percentage and average absorbance of P2X3 receptor were decreased in turn.Compared with CCI group,the mechanical stimulation constraining thresholds of rats were increased,and the positive cells percentage and average absorbance of P2X3 receptor were decreased in the non-MC-OECs group.Compared with non-MC-OECs group,the mechanical stimulation constraining thresholds of rats were increased,the positive cells percentage and average absorbance of P2X3 receptor were decreased in the MC-OECs group.The difference between groups was statistically significant.Conclusion:Microencapsulated OECs transplantation can better alleviate chronic peripheral nerve injury pain and reduce the expression level of P2X3 receptor than non-MC-OECs on the 20 th day after surgery.
