孕期双酚A暴露对子代雄性大鼠生殖发育的影响 被引量:4

摘要 目的:研究孕期双酚A(BPA)暴露对子代雄性大鼠生殖发育的影响。方法:将15只雄性SD大鼠与30只雌性SD大鼠合笼,以次日晨起发现阴栓为妊娠第0天(GD0)。以受孕雌鼠为实验对象,随机分为3组。受试组从GD5开始分别给予不同剂量BPA(33.3 mg/kg、100 mg/kg)玉米油溶液灌胃,连续染毒15 d;对照组给予相同体积的玉米油溶液,各组灌胃体积为5 mg/kg。子代雄性大鼠生后正常喂养至第10周。测定子代雄性大鼠的体重、肛殖距,将子代雄性大鼠乙醚麻醉后取腹主动脉血检测氧化应激指标,测量脏器系数,检测精子数、精子活动百分数等指标。结果:受试组子代雄性大鼠生后第4周至第10周体重与对照组相比差异均有统计学意义(P<0.05);附睾系数比较差异有统计学意义(P<0.01),肝脏系数、肾脏系数、睾丸系数及肛殖距比较差异均无统计学意义(P>0.05);精子活动百分数比较差异有统计学意义(P<0.01);高剂量BPA暴露组精子数明显低于对照组和低剂量BPA暴露组(P<0.01)。与对照组比较,受试组SOD活性下降,高剂量暴露组MDA、H2O2的含量增加(P<0.01)。结论:孕期BPA暴露可影响子代雄性大鼠生长发育水平,对生殖系统产生损伤,造成脂质过氧化损伤,抗氧化酶活性下降,破坏机体抗氧化系统平衡,从而降低对机体的保护能力。 Objective:To study the effect of bisphenol A(BPA)exposure during pregnancy on the reproductive development of offspring male rats.Methods:A total of 15 SD male rats and 30 female rats were conceived in a cage.Pregnant rats were randomly divided into three groups.From gestation days(GD)5-21,the test group was orally given different doses of BPA(33.3 mg/kg,100 mg/kg)in corn oil solution,while the control group was orally given equal volume of corn oil.The gavage volume of each group was 5 mg/kg.The weight and anogenital distance of offspring male rats were detected.Oxidative stress indexes of abdominal aorta blood,viscera coefficient,sperm count,and the percentage of sperm motility were detected after offspring male rats were sacrificed at loth week after birth.Results:There were a statistically significant difference in weight at 4-10 weeks,epididymal visceral coefficient and the percentage of sperm motility between the test group and the control group(P<0.01).The sperm count in 100 mg/kg BPA group was significantly lower than that in 33.3 mg/kg BPA group and control group.The activity of antioxidant enzyme SOD decreased in the test group and the contents of MDA and H2O2 increased in 100 mg/kg BPA group compared with the control group(P<0.01).Conclusions:BPA exposure during pregnancy can affect the growth and decrease development of offspring male rats and damages the reproductive system.BPA exposure can cause lipid peroxidation damage,antioxidant enzyme activity,and destroy antioxidant system balance,thus reduce the protection of the body ability.
