三种线栓制备大鼠MCAO模型的比较 被引量:1

摘要 目的:研究三种尼龙线栓对栓塞法制备大鼠大脑中动脉栓塞(middle cerebral artery occlusion,MCAO)模型的影响,并探讨其意义。方法:SD大鼠随机分为自制线栓组,A4级线栓组,A5级线栓组3个组,经颈外动脉插线制备MCAO模型,观察造模手术操作时间、造模成功率、成模大鼠24 h死亡率、脑梗死体积。结果:自制线栓组插栓平均用时(8.23±2.73)min、A4级线栓组(5.40±1.44)min、A5级线栓组(2.70±0.78)min,组间比较差异有统计学意义(P<0.05);自制线栓组、A4级线栓组、A5级线栓组的成模率分别为62.5%、88.9%、97.9%,组间比较差异有统计学意义(P<0.05);自制线栓组、A4级线栓组、A5级线栓组成模大鼠的24 h死亡率分别为6.7%,5.0%,2.2%,组间比较差异无统计学意义(P>0.05);自制线栓组、A4级线栓组、A5级线栓组脑梗死体积百分比分别为(18.82±12.64)%、(20.59±7.35)%、(35.01±8.00)%,组间比较差异有统计学意义(P<0.05),其中A5级线栓组高于其他两组(P<0.05),自制线栓组梗死体积的稳定性较差。结论:A5级线栓组插线用时少、成模率高、脑梗死体积大且一致,能明显提高造模的稳定性,将线栓对MCAO模型制备的影响降到更低。 Objective:To detect the influence of three types of nylon thread on the embolism method in the preparation of rat MCAO model,and discuss its significance.Methods:SD rats were randomly divided into three groups:homemade line group,A4 plug group and A5 plug group.The thread of MCAO model was pluged from external carotid artery.The plugging operation time,the success rate,the mortality rate after 24 hours and the infarct volume were observed.Results:The average time of plugging was(8.23±2.73)mins in homemade line group,(5.40±1.44)mins in A4 plug group,and(2.70±0.78)mins in A5 plug group(P<0.05).The success rate of establishment was 62.50%in homemade line group,88.9%in A4 plug group and 97.9%in A5 plug group(P<0.05).The mortality rates were 6.7%,5.0%and 2.2%respectively(P>0.05).The average infarction volumes were(18.82±12.64)%,(20.59±7.35)%and(35.01±8.00)%respectively(P<0.05),in which the volume in A5 plug group was higher than those in the other two groups(P<0.05)and the volume in homemade line group had poor stability.Conclusion:A5 plug in the preparation of rat MCAO model has less operation time,higher success rate and more consistent and higher infarct volume,which can greatly improve the stability of the MCAO model.
