摘要
目的建立黑老虎中12-乙酰氧基黑老虎酸和开环新五味子酸A的含量测定方法。方法采用超声法提取药材,色谱柱为Atlantis T3-C_(18)柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.1%磷酸水溶液(80∶20),检测波长:210 nm,流速1.0 mL/min,柱温为30℃。结果12-乙酰氧基黑老虎酸进样量在0.0157~0.3140μg与峰面积值呈现良好的线性关系(r=0.9998),平均回收率为100.4%,相对标准差(RSD)为2.58%(n=6);开环新五味子酸A进样量在0.0848~1.6960μg与峰面积值呈现良好的线性关系(r=0.9999),平均回收率为101.1%,RSD为1.21%(n=6)。结论该方法准确、简便、重复性好,可用于黑老虎药材中12-乙酰氧基黑老虎酸和开环新五味子酸A的含量测定。
Objective To establish a method for the determination of 12-acetoxycoccinic acid and seco-neokadsuranic acid A in root of Kadsura coccinea.Methods Ultrasonic method was used to extract the herbs.HPLC chromatographic conditions included the Atlantis T3-C18(4.6 mm×250 mm,5μm)column,using acetonitrile-0.1%phosphoric acid solution(80∶20)as the mobile phase at a flow rate of 1.0 mL/min.The detection wavelength was set at 310 nm and the column temperature was maintained at 30℃.Results The calibration curve of 12-acetoxycoccinic acid was in good linearity over the range of 0.0157-0.3140μg(r=0.9998).The average recovery was 100.4%with RSD of 2.58%(n=6).And the calibration curve of seco-neokadsuranic acid A was in good linearity over the range of 0.0848-1.6960μg(r=0.9999).The average recovery was 101.1%with RSD of 1.21%(n=6).Conclusion The method is accurate,simple,reproducible,and can be used to determine the content of 12-acetoxycoccinic acid and seco-neokadsuranic acid A in the root of Kadsura coccinea.
