探讨免疫组化检测脑胶质瘤IDH1及应用 被引量:1

摘要 目的胶质瘤中异柠檬酸脱氢酶1(Isocitrate dehydrogenase 1,IDH1)不同检测方法的比较,并探讨IDH1突变对胶质瘤临床诊断的意义。方法搜集胶质瘤患者手术切除送检标本,采用ventana Bench Mark ULTRA全自动免疫组化染色法和手工免疫组化SP染色法检测胶质瘤IDH1的表达。结果全自动免疫组化染色法染色强度强、着色均匀、阳性定位准确、背景清晰,IDH1表达阴性6例,阳性7例;手工免疫组化SP染色染色强度较弱、着色欠均匀、阳性定位不准确、背景不清晰,IDH1表达阴性6例,阳性7例。手工染色与全自动免疫组化染色结果一致。结论在IDH1免疫组化检测中,全自动免疫组化仪染色结果较手工免疫组化染色阳性表达定位准确、着色均匀、背景清晰、无非特异染色,操作简单、运行时间短,并且准确率高。此外IDH1突变阳性的肿瘤细胞并没有随着肿瘤级别的进展而增加,不呈正相关的关系。 Objective To compare the different detection methods of Isocitrate dehydrogenase 1,IDH1 in gliomas and explore the significance of IDH1 mutation in clinical diagnosis of glioma.Methods The specimens of patients with glioma were collected for surgical resection,and the expression of IDH1 in gliomas was detected by using ventana Bench Mark ULTRA automated immunohistochemical staining and manual immunohistochemical SP staining.Results There were 6 cases with negative expression of IDHl and 7 cases with pos itive expres sion of IDHl by immunohistochemical staining with strong staining intensity,uniform staining,accurate positive location and clear background;There were 6 cases with negative expression of IDHl and 7 cases with positive staining of IDHl by manual immunohistochemical SP staining with weak staining intensity,less uniform staining,inaccurate positive location,and unclear background.Manual staining was consistent with full-automatic immunohistochemical staining.Conclusion In the detection of IDHl immunohistochemical staining,compared with manual immunohistochemical staining,the result of automated immunohistochemical staining has accurate positive location,uniform staining,clear background,and there is no non-specific staining.The operation is simple with short running time and high accuracy.In addition,IDHl mutation positive tumor cells did not increase with the progression of tumor grades.There was no positive correlation.
