摘要
目的:观察决明子蒽醌苷对糖尿病大鼠肾损伤保护作用。方法:糖尿病大鼠模型随机分为糖尿病组、决明子蒽醌苷组(0.4g/kg)及雷公藤甲素组(0.4mg/kg),另设正常对照组(n=6),药物或同体积生理盐水灌胃8周。留取24h尿液,采用酶免法检测各组尿KIM-1、β_2-MG含量;放免法检测肾组织内肾素、血管紧张素(AngⅡ)含量。结果:糖尿病组大鼠尿中KIM-1、β_2-MG、肾素、血管紧张素(AngⅡ)含量明显高于正常对照组(P<0.05),决明子蒽醌苷组尿中KIM-1、β_2-MG、肾素、血管紧张素(AngⅡ)含量显著低于糖尿病组(P<0.05),决明子蒽醌苷组与雷公藤甲素组无明显差异(P>0.05)。结论:决明子蒽醌苷对糖尿病大鼠肾损伤有保护作用,该作用与其抑制肾素、AngⅡ表达,降低尿中KIM-1和β_2-MG含量有关。
Objective:To observe the protective effect of cassia glycosides on renal injury in diabetic rats.Methods:Diabetic rat models were randomly divided into the diabetes group,the cassia glycoside group(0.4g/kg)and the triptolide group(0.4mg/kg).In addition,the normal control group(n=6)was established,and the medicine or the same volume of saline was intragastrically administered for 8 weeks.24 hours urine was collected,and the urinary KIM-1 andβ2-MG levels in each group were detected by enzyme-free method.The content of renin and angiotensin(AngII)in the renal tissue was detected by radioimmunoassay.Results:The levels of KIM-1,β2-MG,renin and angiotensin II in urinen the diabetes group were significantly higher than those in the normal control group(P<0.05).The content of KIM-1,β2-MG,renin and angiotensin II in urine in the cassia glycoside group was significantly lower than those in the diabetes group(P<0.05).There was no significant difference between the cassia glycoside group and the triptolide group(P>0.05).Conclusion:Cassia doside has a protective effect on renal damage in diabetic rats.Because cassia glycoside can inhibit renin,AngII expression,and reduce urinary KIM-1 andβ2-MG content.
