摘要
目的通过建立动物模型,研究微小RNA-21在胰岛素抵抗(IR)和糖尿病(DM)伴发非酒精性脂肪肝(NAFLD)发病中的作用。方法选取同窝小鼠等量分为三组:胰岛素抵抗(IR)组、糖尿病(DM)组和对照组;记录小鼠体质量,腹腔糖耐量实验,确定糖代谢异常,行肝脏病理切片检查,检测空腹血糖和肝脏组织miR-21表达改变。结果实验小鼠分别饲养至28天、56天,IR组和DM组与对照组相比,其体质量均有明显升高的趋势(P<0.05);实验小鼠分别饲养至28天、56天,IR组和DM组与对照组相比,其肝脏组织miR-21表达水平有明显下降的趋势(P<0.05)。结论微小RNA-21于非酒精性脂肪肝病的作用机制有着十分密切的联系,miR-21可以作为生物标志物应用于临床上对非酒精性脂肪肝病的诊断和治疗。
Objective To explore the role of microRNA-21(miRNA-21)in the pathogenesis of insulin resistance(IR)and diabetes(DM)with non-alcoholic fatty liver(NAFLD)in animal models.Methods The same nest mice were divided into three groups:the insulin resistance(IR)group,the diabetes(DM)group and the control group;the mice body mass,the abdominal sugar tolerance test,the glucose metabolism abnormality,the liver pathological section examination,the changes of fasting blood glucose and miRNA-21 expression in liver tissue were detected.Results The experimental mice were fed for 28 d and 56 d respectively.The body weight of IR group and DM group was significantly higher than that of the control group(P<0.05).The experimental mice were fed for 28 d and 56 d respectively.The miRNA-21 expression level in the liver tissue was significantly decreased(P<0.05)compared with the control group and the IR group and the DM group.Conclusion The mechanism of miRNA-21 is closely related to the mechanism of NAFLD.MiRNA-21 can be used as a biomarker in the diagnosis and treatment of NAFLD.
