正交试验优选红花注射液药渣中亚精胺的提取工艺及其含量测定 被引量:1

摘要 本文对红花注射液药渣中的亚精胺成分进行了提取工艺研究及含量测定。运用已建立的红花中4个亚精胺高效液相色谱(HPLC)含量测定方法,以4个亚精胺含量测定值为指标,考察提取溶剂、提取方法、料液比、提取时间及提取次数5个因素对红花注射液药渣中4个亚精胺总提取率的影响。根据单因素和正交试验分析确定红花注射液药渣中4个亚精胺的最佳提取工艺条件为:35倍量的无水甲醇热回流提取3次,每次提取2 h,得到红花注射液药渣中4个亚精胺化合物总含量为(2.894±0.011)mg·g-1,该提取工艺较之前建立的HPLC含量测定方法所提取的亚精胺总含量高出36.38%。本文首次对红花注射液药渣中4个亚精胺的提取工艺及其含量测定进行研究,结果证实了红花中的亚精胺类成分并未被水提工艺所提取出来,而是残留在红花注射液药渣中,为药渣资源合理和高效利用提供参考。 In this paper,the extraction process of coumaroylspermidines in safflower injection dregs and its content determination were studied.Based on the established high performance liquid chromatography(HPLC)determination method of coumaroylspermidines in safflower,take the content of four coumaroylspermidines as assessment index,effects of extraction solvent,extraction method,solid-liquid ratio,extraction time and extraction times on the total extraction rate of four coumaroylspermidines in safflower injection dregs were investigated.According to the single factor and orthogonal test analysis,the optimum extraction process of four coumaroylspermidines in safflower injection dregs was determined as follows:35 times the amount of anhydrous methanol was heat reflux three times for 2 hours,the total content of coumaroylspermidines in safflower injection dregs was(2.894±0.011)mg·g-1,which was higher than the total content of coumaroylspermidines extracted by the HPLC method 36.38%.The extraction process of four coumaroylspermidines in safflower injection dregs and its content determination were studied in this paper for the first time.The results confirmed that the coumaroylspermidines in safflower had not been extracted by water extraction,but remained in safflower injection dregs,providing a reference for rational and efficient use of dreg resources.
