激素处理对NtQPT干涉表达烟草种子萌发及生长影响研究 被引量:1

摘要 以超量表达和干涉表达QPT转基因烟草种子及烟苗为材料,通过不同激素处理,探讨激素与QPT协同调节种子萌发、形态生长及激素应答理化指标变化的作用。结果表明,与野生型植株(WT)和超量表达植株(QPT)相比,低浓度(5mg/L)6-BA能显著提高干涉植株(QPTi)种子发芽率、发芽势和发芽指数等种子萌发相关指标;而低浓度(5mg/L)ABA会抑制种子的萌发,与QPTi植株相比,ABA处理对WT和QPT烟株种子萌发抑制率较高。另外,以高浓度(40mg/L)6-BA或20mg/L ABA处理烟苗均能抑制转基因与非转基因植株株高、叶宽、叶长、茎围等形态指标的生长,6-BA对QPT和WT的叶长指标抑制最为显著,分别为15.59%和33.34%,ABA则对WT和QPTi的株高指标抑制最为明显。此外,高浓度的激素处理也同样会引起3种烟草植株SOD酶活及叶绿素a/b、类胡萝卜素及总叶绿素含量的差异变化。 With over-expression and interference expression of QPT transgenic tobacco seeds and tobacco seedlings as materials,the effects of hormone and QPT on synergistic regulation of seed germination,morphological growth and changes in physicochemical indexes of hormone response were discussed through different hormone treatments.The results showed that,compared with wild type plants(WT)and over-expression plants(QPT),low concentration(5 mg/L)6-BA could significantly improve germination related indexes such as seed germination rate,germination potential and germination index of intervention plants(QPTi).However,low concentration(5 mg/L)ABA inhibited seed germination.Compared with QPTi plants,ABA treatment had higher inhibition rate for WT and QPT tobacco seed germination.In addition,tobacco seedlings treated with high concentration(40 mg/L)6-BA or 20 mg/L ABA could inhibit the growth of morphological indexes such as plant height,leaf width,leaf length and stem circumference of transgenic and non-transgenic plants.6-BA had the most significant inhibition on leaf length index of QPT and WT which were 15.59%and 33.34%respectively.ABA had the most obvious inhibition on plant height indexes of WT and QPTi.In addition,high concentration of hormones can also cause differences in SOD activity,chlorophyll a/b,carotenoid and total chlorophyll content in three tobacco plants.
