摘要
为探究酸枣种子结构、萌发过程以及影响种子萌发的因素,本实验采用Box-Behnken响应面法设计三因素三水平正交试验,建立各因素与响应值之间的数学模型;并利用石蜡切片技术观察酸枣种子的胚结构和形态变化。结果表明:酸枣种子在赤霉素浓度480mg/L,温度30℃,光照24h的环境下发芽率最佳;种子发育过程的显微结构显示,胚根的分生细胞显著伸长,子叶、胚乳细胞中的内含物质明显消耗,同时伴随着胚芽、胚轴细胞的逐渐伸长生长。
In order to explore the Jujube seed structure,germination process and the factors influencing seed germination,this experiment used box-behnken response surface method to design three-factor three-level orthogonal test,and established a mathematical model between each factor and response value.The structure and morphological changes of the embryo of Jujube seed were observed by paraffin section technique.The results showed that the germination rate of Ziziphus jujuba seeds was the highest under the condition of 480 mg/L gibberellic acid,T=30℃,light for 24 h.The microstructure of the seed development showed that the meristematic cells of radicle was significantly elongated,the contents of the cotyledon and endosperm cells were obviously consumed,at the same time,it was accompanied by the gradual elongation and growth of germ and cotyledon cells.
