摘要
目的:探讨高血压阴虚阳亢证代谢产物谱及其相互作用。方法:从中国知网、万方数据库、维普数据库、Embase、Pub Med数据库中分别检索高血压阴虚阳亢证患者血浆代谢组学研究,对其纳入研究代谢产物进行汇总。信号通路分析采用KEGG,代谢产物分子注释、相关酶或转运蛋白及其相关性质分析采用HMDB,代谢产物路径可视化采用Met PA网络软件。结果:共有5篇文献纳入研究,16个代谢产物,主要参与了34条代谢通路。通过通路拓扑分析选出了10条P<0.05的代谢通路作为高血压阴虚阳亢证的代谢通路,分别是:氨酰t RNA生物合成,缬氨酸、亮氨酸和异亮氨酸生物合成,甘氨酸、丝氨酸和苏氨酸代谢,脂肪酸生物合成,丙氨酸、天门冬氨酸和谷氨酸代谢,精氨酸和脯氨酸代谢,泛酸和辅酶A生物合成,丙酸代谢,缬氨酸、亮氨酸和异亮氨酸降解,乙醛酸和二羧酸盐代谢。结论:高血压阴虚阳亢证的病理机制涉及糖类、脂肪、蛋白质、维生素及血管活性物质的生物代谢,参与活性氧簇介导的血管内皮损伤,及血管炎症反应。本研究从分子水平阐述肝阳上亢证的发病机制,为其诊断及与其他证型之间的鉴别诊断提供基础,也为发现药物作用靶点提供依据,进一步指导两证中医立法选方。
Objective:The metabolite profiles and their interaction in patients with hypertension of the Yinxu Yangkang syndrome(阴虚阳亢证)were analyzed.Methods:The research of hypertension of the Yinxu Yangkang syndrome was searched from CNKI,Wanfang database,VIP database,Embase and PubMed.The metabolic products were summarized.Signal channel was analyzed by KEGG.Metabolite molecular annotation,related enzymes or transporters and related properties were analyzed by HMBD.Metabolic product path visualization was analyzed by MetPA.Results:5 articles were included in this study.16 metabolites were involved in 34 metabolic pathways.10 metabolic pathways of P<0.05 were selected as pathway in patients with hypertension of the Yinxu Yangkang syndrome by pathway topological analysis,which included aminoacyl-tRNA biosynthesis,valine with leucine and isoleucine biosynthesis,glycine with serine and threonine metabolism,fatty acid biosynthesis,alanine with aspartate and glutamate metabolism,arginine and proline metabolism,pantothenate and CoA biosynthesis,propanoate metabolism,valine with leucine and isoleucine degradation,glyoxylate and dicarboxylate metabolism.Conclusion:The pathological mechanism of hypertension of the Yinxu Yangkang syndrome is related to biological metabolism of sugar,fat,protein,vitamin and vasoactive substance,participates in the vascular endothelial injury mediated by reactive oxygen species,and the vascular inflammatory reaction.The pathogenesis of the Ganyang Shangkang syndrome(肝阳上亢证)was expounded from molecular level.The study provides the basis for the diagnosis and differential diagnosis among other syndromes,discovery of drug targets,and further guided the TCM prescription of two syndromes.
