高温热浪与臭氧交互作用对高血压大鼠影响的实验研究 被引量:2

摘要 通过模拟高温热浪与臭氧协同暴露的实验研究,探讨高温热浪与臭氧交互作用对高血压大鼠的影响。应用人工气候环境暴露仓(Shanghai-METAS)模拟石家庄市2017年7月7日1时~10日23时实际气温和臭氧污染。将健康大鼠和高血压大鼠各自分为对照组和实验组(高温热浪与臭氧协同暴露组),每组各6只大鼠。将实验组放入暴露仓内,进行高温热浪与臭氧协同暴露,3 d后取出,采用腹主动脉终末采血。测量各组大鼠血浆中生化指标的含量,包括炎性因子[白介素-6(IL-6)、C-反应蛋白(CRP)、可溶性细胞间粘附因子-1(sICAM-1)、肿瘤坏死因子-α(TNF-α)]、血管舒缩因子[内皮素1(ET-1)、血管紧张素-Ⅱ(Ang-Ⅱ)、NO]、心血管危险因素[低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、甘油三酯(TG)、总胆固醇(TC)]、氧化应激和血栓形成因子[丙二醇(MDA)、总超氧化物歧化酶(SOD)、纤溶酶原激活物抑制因子1(PAI-1)、D-二聚体(D2D)]。经高温热浪与臭氧协同暴露后健康大鼠和高血压大鼠肛温、血压升高,而心率降低。高血压大鼠和健康大鼠体内的IL-6、CRP、sICAM-1、TNF-α含量升高。高血压大鼠和健康大鼠的ET-1、Ang-Ⅱ、NO含量升高。高血压大鼠和健康大鼠LDL-C、TG、TC升高,而HDL-C降低。高血压大鼠和健康大鼠MDA、PAI-1、D2D含量升高,SOD降低。高血压大鼠在高温热浪与臭氧的协同暴露过程中更易受高温热浪与臭氧的影响。 In order to investigate the interaction between heat wave and ozone on hypertensive rats,heat wave and ozone pollution were simulated in this study.Heat wave and ozone pollution were simulated on the Shanghai Meteorological and Environmental Animal Exposure System on the basis of the heat wave and ozone pollution data collected in July 2017 in Shijiazhuang,China.Experiments were commenced at 0 o clock on 7 July 2017 and ended at 24 o clock on 10 July.Healthy rats and hypertensive rats were randomly divided into a control group and an experimental group(heat wave and ozone group)respectively,each group had 6 rats.The rats in the experimental groups were exposed to heat wave plus ozone exposure,after three days,the experimental groups were removed from the chamber and blood was collected from the abdominal aorta by vacuum blood collection.The levels of inflammatory factors[interleukin-6(IL-6),C-reactive protein(CRP),soluble intercellular adhesion factor-1(sICAM-1)and tumor necrosis factor alpha(TNF-α)],vasomotion factors[endothelin(ET-1),angiotensin(Ang-II)and NO],cardiovascular risk factors[low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),triglyceride(TG)and total cholesterol(TC)],oxidative stress and thrombosis factors[malondialdehyde(MDA),superoxide dismutase(SOD),plasminogen activator inhibitor 1(PAI-1)and D-dimer(D2D)]were measured in each rat.After the exposure of heat wave plus ozone,rectal temperature,systolic pressure of both rats increased,except for the decreased level of heart rate.There were increased level of inflammatory factors IL-6,CRP,sICAM-1,TNF-αand vasomotion factors ET-1,Ang-Ⅱ,NO after exposure,except for the decreased levels of HDL-C and SOD.Heat wave and ozone increased cardiovascular risk factors LDL-C,TG,TC and oxidative stress and thrombosis factors MDA,PAI-1,D2D.Hypertensive rats were more vulnerable when suffering heat wave and ozone.
