平顶山地区奶牛隐孢子虫种类鉴定及遗传进化分析 被引量:1

摘要 为了阐明河南省平顶山地区奶牛隐孢子虫的流行情况、种类分布与遗传进化分析情况,应用饱和蔗糖漂浮法对平顶山地区233份奶牛粪便进行隐孢子虫病流行情况调查,同时应用分子生物学技术分析隐孢子虫分离株的种类及遗传进化关系。结果显示,223份奶牛粪便中有12份为隐孢子虫阳性,阳性率为5.38%,不同年龄阶段和奶牛场的奶牛隐孢子虫阳性率具有显著差异(P<0.05)。序列比对与遗传进化分析结果显示,平顶山地区12株奶牛隐孢子虫分离株的18S r RNA基因与Genbank中收录的牛隐孢子虫(Cryptosporidium bovis)同源性最高,且两者在系统进化树上为同一大分支,说明此次分离的奶牛隐孢子虫分离株均为牛隐孢子虫。本次研究结果为河南平顶山地区奶牛隐孢子虫的分子流行病学与防治研究奠定了理论基础。 The purposes of the present study were to understand the infection statues and phylogenetic analysis of Cryptosporidium from Pingdingshan in Henan province.Totally 223 fecal samples were collected from Pingdingshan to investigate the prevalence of Cryptosporidium using saturated sucrose floating method,the results showed high infection rate(5.38%)of Cryptosporidium was detected in this study,the infection rates of different developing stages and farms of cattle had obvious differences(P<0.05).Sequence and phylogenetic analysis revealed that 12 Cryptosporidium positive samples showed the highest homology with Cryptosporidium bovis available in Genbank compared with others,and they clustered into the same one branch.The results of the present study would provide theoretical principles for the control and molecular epidemiology of Cryptosporidium in Pingdingshan from Henan province.
