厚朴中蛋白质提取方法的优化 被引量:2

摘要 以湖北产地厚朴为原料,对其蛋白质提取方法进行优化,以满足蛋白质电泳的要求。比较3种不同蛋白质提取方法的提取率,选取超声破碎仪提取法进一步优化,通过单因素试验和正交试验确定最佳条件为超声时间30 min、超声时间间隔6 s、超声功率250 W,得到的蛋白质浓度为930.2μg/m L,研究表明此法可为厚朴的蛋白质电泳指纹图谱研究奠定基础。 The protein extraction method in Magnolia officinalis from Hubei was optimized to meet the requirements of protein electrophoresis.By comparing the protein extraction yield of three different methods,ultrasonic extraction method was selected for further optimization.The optimum conditions were determined by single factor test and orthogonal test,which were as follows:the ultrasonic time 30 min,ultrasonic interval 6 s,ultrasonic power 250 W.Under this condition,the concentration of protein was 930.2μg/mL.The results showed that the method could lay a foundation for the study of protein fingerprint of Magnolia officinalis.
