分心木黄酮超声-微波协同提取及抗氧化性研究 被引量:24

Study on Diaphragma juglandis fructus Flavonoids Ultrasound-microwave Synergistic Extraction Process and Its Antioxidant Activity

摘要 在单因素试验基础上应用响应面分析法(Box-Benhnken)对核桃分心木中黄酮的提取工艺进行优化并研究其体外抗氧化活性。试验结果表明:最佳提取条件为乙醇浓度为50%,料液比为1∶25.6(g/m L),超声功率为261.8 W,微波功率150 W,提取温度为51.6℃,提取时间为8.5 min,分心木黄酮得率为5.17%。抗氧化活性试验结果表明当分心木黄酮浓度为2 mg/m L时,对·OH和O2-·的最大清除率分别为66.2%、36.9%,而对DPPH·的清除率在分心木黄酮浓度为0.07 mg/m L时即达到70.2%,并且具有很强的还原能力。 Diaphragma juglandis fructus as raw material,single factor and response surface analysis the extraction process and antioxidant activity of flavonoid were studied.The results showed that the optimal conditions for the extraction of flavonoids from Diaphragma juglandis fructus were conditions of ethanol concentration of 50%,solid-liquid ratio was 1∶25.6(g/mL),ultrasonic power was 261.8 W,microwave power was 150 W,extraction temperature was 51.6℃and extraction time was 8.5 min.Under these conditions,the extraction effect of flavonoids from Diaphragma juglandis fructus was the best,and the content reached 5.17%.The max clearance rate on·OH,O2-·were 66.2%,36.9%when the concentration of Diaphragma juglandis fructus flavonoids was 2 mg/mL.The scavenging rate on DPPH·was 70.2%when the concentration was 0.07 mg/mL and showed a strong reducing powerhas strong antioxidant activity in vitro.
