乳腺癌内TNFR1的表达及与临床病理因素的相关性

摘要 目的:检测乳腺癌组织内TNFR1的表达,并分析与临床病理因素的相关性。方法:选取2015年9月-2016年1月牡丹江肿瘤医院乳腺外科手术切除的30例乳腺癌标本为试验组,距离肿瘤边缘1 cm处的癌旁组织为对照组。应用Real-time PCR和免疫组织化学技术分别检测TNFR1 mRNA和蛋白的表达。结果:Real-time PCR检测显示,试验组TNFR1 mRNA的表达量为(0.07±0.03),高于对照组的(0.03±0.01),比较差异有统计学意义(P<0.05);免疫组织化学染色显示,试验组TNFR1蛋白呈棕黄色,主要分布于细胞质内,呈弥漫性或局灶性分布,对照组TNFR1蛋白表达不明显;试验组TNFR1蛋白表达为(145.36±3.46),明显高于对照组的(119.69±14.28),比较差异有统计学意义(P<0.05);乳腺癌内TNFR1蛋白的表达与患者年龄、肿瘤大小均无关(P>0.05),而与淋巴结转移及临床分期均密切相关(P<0.05)。结论:乳腺癌内TNFR1的表达可作为判断乳腺癌复发转移的重要标记物。 To detect TNFR1expression in breast cancer tissues and analyze its correlation with clinicopathological factors.Method:A total of30breast cancer specimens from breast surgery in Mudanjiang Cancer Hospital from September2015to January2016were selected as experimental group,and the adjacent tissues1cm from the edge of the tumor as control group.TNFR1mRNA and protein expression were detected by Real-time PCR and immunohistological staining respectively.Result:Real-time PCR detection showed that the expression of TNFR1mRNA in experimental group was(0.07±0.03),which was higher than(0.03±0.01)in control group,the difference was statistically significant(P<0.05).Immunohistochemical staining showed that the TNFR1protein in experimental group was brown and yellow,mainly distributed in the cytoplasm,showing diffuse or focal distribution,and the expression of TNFR1protein in control group was not obvious.The expression of TNFR1protein in experimental group was(145.36±3.46),which was significantly higher than(119.69±14.28)in control group,the differences was statistically significant(P<0.05).The expression of TNFR1protein in breast cancer was not associated with age and tumor size(P>0.05),but was closely related to lymph node metastasis and clinical stage(P<0.05).Conclusion:The expression of TNFR1in breast cancer can be used as an important marker for the recurrence and metastasis of breast cancer.
