低温胁迫下油棕WRKY转录因子基因的表达特性分析 被引量:11

摘要 【目的】研究低温胁迫下WRKY转录因子基因表达特性,并分析其与油棕抗寒性的相关性,为阐明WRKY转录因子在油棕生长和低温响应机制中的功能和作用提供理论参考。【方法】以油棕品种XJS30和SJ64为材料,对其进行低温驯化处理(0 h、1 d和7 d)及冷处理(10℃4 h、8℃4 h、6℃4 h、4℃4 h和2℃4 h),利用实时荧光定量PCR(qPCR)检测不同处理时间的WRKY1、WRKY7、WRKY22、WRKY40和WRKY55基因表达特性,并分析其与油棕抗寒性的相关性。【结果】WRKY1和WRKY7基因在低温驯化结束时的表达量较冷处理结束时高,说明其在低温驯化过程中对XJS30的抗寒性构成发挥调控作用。WRKY22、WRKY40和WRKY55基因均在XJS30冷处理中表达量最高,说明其在冷处理过程中对XJS30的抗寒性发挥调控作用。WRKY1基因在低温驯化结束时的表达量较冷处理结束时低,说明其在冷处理过程中对SJ64的抗寒性发挥调控作用。WRKY7基因在低温驯化结束时的相对表达量较冷处理结束时高,说明其在低温驯化过程中对SJ64的抗寒性构成发挥调控作用。【结论】油棕的WRKY1、WRKY7、WRKY22、WRKY40和WRKY55基因均属于低温应激反应型基因。 【Objective】In this study,expression characteristics of WRKY transcription factor genes under cold stress were revealed,its correlation with cold resistance of oil palm was studied and layed a foundation for researching the function of WRKY transcription factor genes in the growth of oil palm and response mechanisms of cold stress.【Method】In this experiment,oil palm varieties XJS30 and SJ64 were used for analysis of the expression characteristics of WRKY1,WRKY7,WRKY22,WRKY40 and WRKY55 genes in response to various cold treatments by real-time quantitative fluorescence PCR(qPCR),in order to reveal the function of WRKY genes in the process of oil palm in response to low temperature.The materials were treated by low temperature acclimation(0 h,1 d and 7 d)and cold treatment(10℃4 h,8℃4 h,6℃4 h,4℃4 h and 2℃4 h).【Result】The expression levels of WRKY1 and WRKY7 genes at the end of low temperature acclimation were higher than those at the end of cold treatment,the two genes played a regulatory role in the cold resistance of XJS30 during low temperature acclimation.The expression levels of WRKY22,WRKY40 and WRKY55 genes were the highest during cold treatment in XJS30,they played a regulatory role in the cold resistance of XJS30 during cold treatment.The expression levels of WRKY1 gene at the end of low temperature acclimation were lower than that at the end of cold treatment,it played a regulatory role in the cold resistance of SJ64 during cold treatment.The expression level of WRKY7 gene at the end of low temperature acclimation was higher than that at the end of cold treatment,it played a regulatory role in the cold resistance of SJ64 during low temperature acclimation.【Conclusion】It is suggested that WRKY1,WRKY7,WRKY22,WRKY40 and WRKY55 in oil palm belong to the cold stress response genes.
