响应面法优化酶解单环刺螠内脏多肽的工艺研究 被引量:5

摘要 以单环刺螠内脏为原料,利用酶解法提取制备单环刺螠内脏多肽。在单因素试验的基础上,根据BoxBehnken的中心组合试验设计原理,运用Design Expert8.0.5数据统计分析软件,采用三因素三水平的响应面分析法,以多肽得率为响应值,研究加酶量、时间、温度对单环刺螠内脏多肽得率的影响,并优化提取工艺。确定酶解法提取单环刺螠内脏多肽的最佳工艺条件为:料液比1∶10(g/mL),加酶量3.22%,酶解时间3.17 h,温度44.3℃,在此条件下水解度可达17.79%。 Viscera polypeptide of Urechis unicinctus was obstained by enzymatic hydrolysis with the viscera of Urechis unicinctus as raw material.The preparation process of viscera polypeptide was optimized on foundation of single factor experiment,according to Box-Behnken central composition design principle,the method of response surface analysis with 3 factors and 3 levels combined with Design Expert8.0.5-statistical data analysis software was adopted with the leaching rate of viscera polypeptide of Urechis unicinctus as the response value.The effect of volume of enzyme,time and temperature on the leaching rate of viscera polypeptide of Urechis unicinctus were studied.According to the above results,the optimum preparation conditions were obtained as follows:solid liquid ration 1∶10(g/mL),volume of enzyme 3.22%,time 3.17 h,temperature 44.33℃.Under these conditions,the leaching rate of viscera polypeptide of Urechis unicinctus reached 17.79%.
