摘要
目的探讨富氢盐水是否能够拮抗顺铂引起的小鼠耳毒性。方法取健康、ABR阈值正常C57小鼠30只,雌雄不限,体重25-30g。随机分成3组,对照组(单纯腹腔注射0.9%生理盐水4mg/kg)、顺铂组(单纯腹腔注射顺铂4mg/kg)、富氢盐水组(腹腔注射顺铂4mg/kg后立即腹腔注射富氢盐水1ml/100g)各10只,分别连续腹腔注射上述液体7天,行听性脑干诱发反应(auditory brainstem response ABR)检测后处死小鼠取耳蜗行扫描电镜观察。结果给药后,富氢盐水组ABR阈值明显低于顺铂组ABR阈值(P<0.001,P=0.000),高于对照组ABR阈值(P<0.001,P=0.000),有统计学意义。扫描电镜结果显示顺铂组耳蜗内、外毛细胞静纤毛损伤缺失程度明显重于富氢盐水组。结论富氢盐水能够有效拮抗顺铂引起的小鼠耳毒性。
Objective To evaluate hydrogen-rich saline on cisplatin-induced ototoxicity in mice.Methods Thirty C57 mice were randomly divided into a normal saline group(control),a cisplatin group and a hydrogen-rich saline group.For saline or cisplatin solution administration,mice were given daily abdominal injections(4 mg/kg)for 7 days.For hydrogen-rich saline administration,mice were given daily abdominal injections of hydrogen-rich saline immediately after cisplatin administration.Hearing thresholds were estimated by auditory brain responses(ABRs),and changes in the cochlea were observed under a scanning electron microscopy.Results ABR thresholds in the hydrogen-rich saline treated group were significantly lower than those treated with cisplatin alone(P<0.001),although still higher than in the control group(P<0.001).Scanning electron microscopy results showed significantly less hair cells damage in the hydrogen-rich saline group compared with the cisplatin group.Conclusion Hydrogen-rich saline could significantly reduce damage in the inner ear induced by cisplatin in mice.
