摘要
采用分光光度法建立几丁质和麦角固醇标准品的标准曲线,对双孢蘑菇(Agaricus bisporus)W192液体发酵菌丝的2种细胞组分(几丁质和麦角固醇)进行测定,建立了双孢蘑菇W192中几丁质和麦角固醇含量与菌丝量的标准曲线。根据该标准曲线,对双孢蘑菇W192麦粒栽培种的几丁质和麦角固醇含量以及相对菌丝量进行了研究。结果表明:不同质量浓度的几丁质和麦角固醇标准品分别与吸光度值之间具有良好的线性关系,决定系数R2分别达到0.9991和0.9996;W192菌丝体中的几丁质和麦角固醇质量浓度分别与菌丝体质量之间也具有较好的线性关系,决定系数R2均达到0.9991;双孢蘑菇W192液体发酵纯菌丝(7d菌龄)的几丁质和麦角固醇含量分别为121.93、6.145μg/mg;两种方法测定的双孢蘑菇W192麦粒栽培种(15d菌龄)的相对菌丝量分别为120.8mg/g(基于几丁质计算)和139.2mg/g(基于麦角固醇计算),并且2种方法均测出双孢蘑菇麦粒栽培种麦麸中的菌丝量超过胚乳中菌丝量的20~40倍。
Standard curves of chitin and ergosterol were established by spectrophotometry and then used to determine chitin and ergosterol content in the liquid mycelium culture of Agaricus bisporus W192.Linear regression was then established between mycelial mass and chitin or ergosterol content in the culture.The results showed that there was a good linear relationship between OD value and chitin/ergosterol at different concentrations,and the coefficient of determination R 2 of chitin and ergosterol were 0.9991 and 0.9996,respectively.There was also a good linear relationship between mycelial mass of submerged culture and the ergosterol and chitin contents therein,and both R 2 were 0.9991.Chitin and ergosterol in submerged fermented mycelia of A.bisporus W192 were 121.93 and 6.145μg/mg,respectively.The mycelial mass of A.bisporus W192 wheat spawn at day 15 was estimated to be 120.8 mg/g based on chitin content and 139.2 mg/g based on ergosterol content.Both algorithms revealed that the mycelial mass in the wheat bran of A.bisporus W192 wheat spawn was 20-40 times greater than that in the endosperm.
