大白菜BrCOX11基因的克隆及表达分析

摘要 为了研究大白菜中细胞色素C氧化酶的功能,以雄性不育系及保持系、抗感根肿病DH系为材料,在对大白菜BrCOX11基因克隆的基础上,利用生物学软件对其编码蛋白进行分析和构建进化树。结果显示,大白菜BrCOX11基因编码区大小为861 bp,所编码的氨基酸序列与NCBI收录的COX11氨基酸序列相似性非常高,属于COX家族;分子进化树分析表明,大白菜BrCOX11与芸薹属、拟南芥属及萝卜属亲缘关系最近,而与棉花属、胡萝卜属和黄瓜属等亲缘关系较远;大白菜BrCOX11蛋白是亲水性蛋白。通过分析BrCOX11基因表达发现,BrCOX11基因的表达量和大白菜的生长时期有关系,在果荚中表达量最高;在根肿菌侵染后的根部组织中表达上调,而在根肿菌侵染后的叶片组织中表达下调;在保持系中的表达量明显高于不育系。 In order to study cytochrome C oxidase’s function in Chinese cabbage,the male sterile lines and maintainer,clubroot resistance and susceptible double haploid lines were used as the experimental material.Based on the cloning of the BrCOX11 gene of Chinese cabbage,the encoding protein was analyzed and the evolutionary tree was constructed.The results showed that the complete open reading frame of BrCOX11 gene contained 861 bp,and the similarity of the amino acid sequences of BrCOX11 with other COX11 was very high,which belonged to the COX family.The molecular phylogenetic tree showed that BrCOX11 was closely related to Brassica,Arabidopsis and radish,while far from the genus of cotton,carrot and cucumber.The BrCOX11 protein was a hydrophilic protein.Through gene expression analysis,its expression was related to the growth period of Chinese cabbage,and showed the highest expression in silique.It was upregulated in root tissues after infection,but decreased in leaf tissue.Its expression in maintainer line was higher than that of male sterile lines.
