肝细胞癌组织核因子E2相关因子3表达变化及意义 被引量:3

Expression and clinical significance of NFE2L3 in hepatocellular carcinoma

摘要 目的探讨核因子E2相关因子3(NFE2L3)在肝细胞癌发生、发展中的作用。方法选取肝细胞癌患者44例,采用免疫组化两步法检测肝癌组织及癌旁正常组织NFE2L3表达,分析肝癌组织NFE2L3表达与患者临床病理参数的关系;分析癌症基因组图谱(TCGA)数据库肝细胞癌数据集中371例份肝癌组织和51例份癌旁正常组织中NFE2L3差异表达情况。结果肝癌组织与癌旁正常组织NFE2L3阳性表达率分别为70. 45%(31/44)、15. 91%(7/44),肝癌组织NFE2L3阳性表达率明显高于癌旁正常组织(P <0. 05)。肝癌组织NFE2L3阳性表达与肿瘤组织分化程度、患者是否合并乙肝相关(P均<0. 01),与患者年龄、性别、是否合并肝硬化、肝功能分级及TNM分期均无关(P均> 0. 05)。TCGA数据库肝细胞癌数据集分析显示,肝癌组织NFE2L3表达显著高于癌旁正常组织,log2FC (肝癌组织与癌旁正常组织差异表达倍数)=0. 750 121 651,Ave Expr (NFE2L3平均表达值)=2. 336 727 847,P <0. 01。结论 NFE2L3可能作为促癌因子参与肝细胞癌的发生、发展。 Objective To investigate the expression and significance of NFE2L3 in hepatocellular carcinoma(HCC).Methods We selected 44 cases of HCC patients,and the expression of NFE2L3 protein in the HCC tissues and adjacent normal tissues was detected by two-step immunohistochemical method.The relationship between expression of NFE2L3 and clinicopathological features of HCC was analyzed.The differential expression of NFE2L3 in 371 HCC tissues and 51 adja-cent tissues from the cancer genome atlas(TCGA)database was also analyzed.Results The positive expression rates of NFE2L3 in the HCC tissues was 70.45%(31/44),which was higher than that of the adjacent tissues[15.91%(7/44)],P<0.05.The expression of NFE2L3 in HCC tissues was related to tumor differentiation and hepatitis B virus infec-tion(P<0.01),but was not related to age,sex,cirrhosis,liver function grade and clinical stage of tumor(all P>0.05).The expression of NFE2L3 in the HCC tissues was significantly higher than that in the adjacent tissues from TCGA(loggFC=0.750 121 651,AveExpr=2.336 727 847,P<0.01).Conclusion NFE2L3 may be involved in the carcino-genesis and development of HCC as an oncogene.
