摘要
目的探讨中药抗氧化剂槲皮素(quercetin,Que)和茶多酚对PC12细胞的毒性及对光动力效应的影响。方法 MTT法测定12.5~100μmol·L^(-1)槲皮素、茶多酚孵育24 h,对PC12细胞的毒性及对PSD-007介导的光动力效应的影响;LDH试剂盒检测细胞培养液上清LDH活性,研究对PC12细胞膜的作用。结果槲皮素6.25μmol·L^(-1)使PC12细胞存活率降低10.7%,50~100μmol·L^(-1)槲皮素使细胞存活率降低约21.5%。茶多酚6.25~25μmol·L^(-1)作用的细胞存活率无明显变化,茶多酚25~100μmol·L^(-1)作用时,随浓度增大,细胞存活率最多降低了33.3%。与PDT组比较,预加入槲皮素12.5、25、50μmol·L^(-1)后再PDT,细胞存活率分别增加了1.166倍、1.134倍、1.129倍,PC12细胞上清中LDH降低了63.0%;茶多酚12.5、25、50μmol·L^(-1)作用时,细胞存活率分别降低了14.0%、8.8%、17.2%, PC12细胞上清中LDH与PDT组相比,无明显变化(P>0.05)。结论在PC12细胞模型上,槲皮素对PSD-007介导的光动力效应具有拮抗作用,茶多酚对PSD-007介导的光动力效应具有协同作用,细胞膜可能是槲皮素保护作用的靶点。
To investigate the cytotoxicity of antioxidant quercetin and tea polyphenols(TP)as well as the effects against photodynamic therapy(PDT)on PC12 cells.Methods After 24 hours’co-incubation at 37℃,the toxicity of 12.5~100μmol·L-1 of quercetin and tea polyphenols on PC12 cells was tested with MTT assays,and their effects against oxidative damages induced by PSD-007 PDT on PC12 cells were measured with MTT assays,also the activity of lactic acid dehydrogenase(LDH)in the upper layer of PC12 culture solution was tested by LDH assay to examine the damage to the cell membrane.Results Compared with control group,the cell survival rate decreased by 10.7%for 6.25μmol·L-1 of quercetin,and decreased by approximately 21.5%for 50~100μmol·L-1 of quercetin.The cell survival rates did not change obviously during 6.25~25μmol·L-1 of TP,but decreased by 33.3%as the concentration of TP increased from 25 to 100μmol·L-1.Compared with PDT group,the cell survival rate increased by 1.166 times,1.134 times and 1.129 times respectively for 12.5,25,50μmol·L-1 of quercetin added pre-PDT,and the activity of LDH all decreased by about 63.0%in supernatant of PC12 cells.The cell survival rate decreased by 14.0%,8.8%,17.2%respectively for 12.5,25 and 50μmol·L-1 of TP,and the activity of LDH in supernatant had no significant changes compared with that of PDT group(P>0.05).Conclusions Quercetin has antagonistic effect on PDT induced by PSD-007,but tea polyphenols has synergistic effect on PC12 cells.Cell membrane may be the protected target of quercetin.
