^(18)F-FLT micro PET/CT显像评估马蔺子素对裸鼠乳腺癌的放射增敏作用 被引量:5

^(18)F-FLT micro PET/CT imaging evaluation of radiosensitization of MDA-MB231 breast cancer in nude mice

摘要 目的利用^(18)F-FLT micro PET/CT显像评估马蔺子素(IR)对裸鼠MDA-MB231乳腺癌的放射增敏作用。方法 32只裸鼠建立MDA-MB231乳腺癌模型,随机分为对照组、IR组、单纯放疗组、IR+放疗组,每组8只。分别在放疗前及放疗后24 h,行^(18)F-FLT micro PET/CT显像,观察4组肿瘤SUV_(max)的变化。免疫组织化学法分析Ki-67的表达情况。结果放疗前,4组显像SUV_(max)差异无统计学意义(P>0.05);放疗后24 h,对照组及IR组SUV_(max)较治疗前显像升高(P<0.05),单纯放疗组和IR+放疗组显像SUV_(max)较治疗前显像下降(P<0.05),且IR+放疗组较单纯放疗组下降更明显(P<0.05);免疫组化结果显示,放疗组和IR+放疗组中Ki-67阳性表达均明显低于对照组(P<0.05)。Ki-67表达与SUV_(max)呈明显正相关(r=0.897,P<0.05)。结论 IR对裸鼠乳腺癌具有放射增敏作用,^(18)F-FLT micro PET/CT可以评估IR对裸鼠乳腺癌的放射增敏作用。 To study 18 F-FLT micro PET/CT imaging for the evaluation of irisquinone(IR)radiosensitizing effect on MDA-MB231 breast cancer in nude mice.Methods The 32 MDA-MB231 breast cancer model in nude mice was established and randomly divided into control group,IR group,radiotherapy group and IR+radiotherapy group.18 F-FLT micro PET/CT imaging was performed before and 24 h after radiotherapy respectively,and the changes of four groups of tumor SUV max were observed.The expression of Ki-67 was analyzed by immunohistochemistry.Results Before radiotherapy,there was no statistically significant difference in SUV max among the four groups(P>0.05).24 h after radiotherapy,SUV max in control group and IR group increased significantly(P<0.05),while SUV max in radiotherapy group and IR+radiotherapy decreased(P<0.05),and IR+radiotherapy group decreased more significantly than radiotherapy group(P<0.05).The immunohistochemical results showed that the positive expression of Ki-67 in radiotherapy group and IR+radiotherapy group was significantly lower than that in control group(P<0.05).The expression of Ki-67 was positively correlated with SUV max(r=0.897,P<0.05).Conclusions IR has radiosensitizing effect on breast cancer in nude mice.18 F-FLT micro PET/CT can evaluate the radiosensitization effect of IR on breast cancer in nude mice.
