12年生转基因毛白杨外源基因转移及其对土壤微生物的影响 被引量:6

摘要 对大田种植12 a的转基因毛白杨(Populus tomentosa)根、茎、叶组织基因组DNA以及土壤总DNA和卡那霉素抗性菌株基因组DNA进行PCR特异性扩增检测,研究了转基因毛白杨对根际土壤可培养微生物数量时空动态的影响。结果表明:大田种植12年后转基因毛白杨的外源基因仍然存在于植株的各组织中;转基因毛白杨林下土壤总DNA和卡那霉素抗性菌株基因组DNA中,均未检测出与预期长度符合的条带;转基因和对照(非转基因毛白杨)林下根际土壤微生物数量无显著差异;随着土层深度的增加转基因与对照根际土壤微生物中细菌和放线菌数量均显著减少,而真菌无明显变化。在水平方向上,转基因与对照根际土壤微生物中真菌和放线菌数量均无显著差异,仅在土壤深度40 cm处,水平方向20 cm与水平方向50 cm处的细菌数量有显著差异。表明,大田种植12年后转基因毛白杨暂未对土壤可培养微生物数量造成显著影响。 PCR amplification was used to detect roots,stems,leaves DNA and the total soil DNA and the genomic DNA of the kanamycin resistant strain of transgenic Populus tomentosa in field for 12 a.The effects of transgenic P.tomentosa in the number of rhizosphere soil microbes were studied,and the effects of transgenic P.tomentosa in the number of rhizosphere soil microbes on spatial and temporal dynamic changes were studied by further in-depth study.It was proved that the exogenous genes of P.tomentosa were still present in the tissues of the plant in field for 12 a.There was no band corresponding to the expected length in total genomic DNA and kanamycin resistance strain genomic DNA of transgenic P.tomentosa.There was no significant difference between transgenic and non-transgenic P.tomentosa in the number of rhizosphere soil microbial.With the increase of soil depth,there was significantly decrease between transgenic and non-transgenic P.tomentosa in the number of bacteria and actinomycetes of rhizosphere soil,but there was no significant change in fungi.There was no significant difference in the number of fungi and actinomycetes in rhizosphere soil of transgenic and non-transgenic P.tomentosa in the horizontal direction.But,at the soil depth of 40 cm,the number of bacteria was significantly reduced from the horizontal direction 20 to 50 cm.The transgenic P.tomentosa had no significant effect on the soil ecosystem after 12 a of field cultivation.
