小鼠开胸前气管内灌注肺制样方法体重依赖灌注量的探讨

Discuss on the Weight-dependent Perfusion Volume of Mice in the Preparation of Lung Samples by Intratracheal Infusion Before Opening the Chest

为探讨小鼠开胸前气管内灌注肺制样方法体重依赖的最适灌注量,通过采用开胸前气管内灌注肺制样并根据体重从0~0.2mL/20g探索灌注量,固定后进行切片HE染色观察固定效果。结果表明在0.2mL/20 g时,肺组织灌注均匀,切片后组织结构清晰,人为假象较少。可见小鼠开胸前气管内灌注肺制样灌注量为0.2mL/20 g可取得良好的组织固定效果。

In order to investigate the weight-dependent optimal perfusion volume of mice in the preparation of lung samples by intratracheal infusion before opening the chest.Samples were prepared by intratracheal infusion before opening the chest and the perfusion volume was probed according to the body weight from 0 to 0.2 mL/20 g.After fixing,the fixation effect was observed by sectioning HE staining.The results show that in 0.2 mL/20g,the lung tissue was evenly perfused,the histological structure was clear after slicing,and there was less artificial artifact.It is concluded that in mice,0.2 mL/20 g perfusion of the preparation of lung samples by intratracheal infusion before opening the chest can achieve good tissue fixation.
