斯鲁菲亚海洋红酵母培养基优化

摘要 [目的]比较不同的碳源、氮源、无机盐对斯鲁菲亚海洋红酵母(Rhodotorula slooffiae)菌株产量的影响,确定培养基最优组分。[方法]通过葡萄糖、酵母浸粉、硫酸铵、磷酸二氢钾4因素3水平的正交试验,确定斯鲁菲亚海洋红酵母的最优培养基。[结果]斯鲁菲亚海洋红酵母的最优培养基各成分质量分数为:葡萄糖2%,酵母浸粉0.75%,硫酸铵0.5%,磷酸二氢钾1%。接种量6%,500 m L三角锥形瓶,装液量100 m L,培养温度30℃,经过48 h培养,菌种量最大可达8.6×10~8CFU/m L,比优化前的发酵培养基产量(6.6×10~8CFU/m L)提升了30%。[结论]培养条件和工艺的优化可以显著提高海洋红酵母菌量和类胡萝卜素含量。 [Objective]The effects of different carbon sources,nitrogen sources and inorganic salts on fermentation yield of marine red yeast(Rhodotorula slooffiae)were compared to determine the optimal medium components.[Method]The optimum fermentation conditions for Rhodotorula slooffiae were determined by 4 factors and 3 levels orthogonal test.[Result]The optimum fermentation conditions for Rhodotorula slooffiae were determined:glucose 2%,yeast extract powder 0.75%,ammonium sulfate 0.5%,monopotassium phosphate 1%,and inoculation amount 6%,temperature 30℃,100 mL medium in 500 mL triangle bottle and fermentation time 48 h.Under above conditions,the cell biomass of marine red yeast can reach 8.6×10 8 cfu/mL,which increased by 30%as compared with the fermentation before optimization.[Conclusion]Optimization of culture conditions and process can significantly increase the amount of marine red yeast and carotenoid content.
