响应面法优化离子液体双水相萃取木瓜蛋白酶 被引量:11

Optimization of Ionic Liquid Aqueous Two-phase Extraction of Papain by Response Surface Methodology

摘要 采用1-乙基/丁基/戊基/辛基-3-甲基咪唑醋酸盐{[C_nmim]Ac(n=2,4,6,8)}与K_2HPO4离子液体双水相体系对木瓜蛋白酶进行萃取分离。用浊点法测定双水相体系的双节线并采用Merchuk方程对数据进行拟合。考察各成相试剂及pH值对木瓜蛋白酶分配行为的影响,利用单因素试验和响应面法确定其离子液体双水相最佳萃取条件为:离子液体[C_8mim]Ac浓度为28.03%,K_2HPO_4浓度为20.08%,pH值为8.55。在该条件下,分配系数可达到11.65。 The papain was extracted and separated using 1-ethyl/butyl/pentyl/octyl-3-methylimidazolium acetate{[Cnmim]Ac(n=2,4,6,8)}and K2HPO4 ionic liquid aqueous two phase system(IL-ATPS).The binodal curve of the aqueous two-phase system(ATPS)was determined using the cloud point method and the data was fitted using the Merchuk equation.The effects of various phase forming reagents and pH on the partitioning behavior of papain were investigated.The optimum extraction conditions for aqueous two-phase extraction(ATPE)were determined by single factor test and response surface methodology:the ionic liquid[C8mim]Ac concentration was 28.03%,The K2HPO4 concentration was 20.08%and the pH value was 8.55.Under this condition,the partition coefficient can reach 11.65.
