燕麦β-葡聚糖对脓毒血症大鼠肠黏膜损伤的保护作用及其机制探讨 被引量:1

摘要 目的探索燕麦β-葡聚糖(Oglu)能否对脓毒血症空肠损伤提供保护。方法选取50只SD大鼠,按随机数字表法分为对照组、脓毒血症组、Oglu组。脓毒血症组和Oglu组以改良盲肠结扎穿孔法复制脓毒血症模型,通过病理检查评估空肠损伤。采用酶联免疫吸附法检测炎症因子白细胞介素-6(IL-6),肿瘤坏死因子-α(TNF-α)和降钙素原,PCR、Western blot检测低氧诱导因子-1(HIF-1α)、β-连环蛋白(β-catenin)、T细胞因子(TCF-4)、基质金属蛋白酶-13(MMP-13)表达的变化。结果 Oglu缓解脓毒血症诱导的空肠损伤,降低脓毒血症大鼠IL-6、TNF-α及降钙素原水平。Oglu组空肠组织中HIF-1α表达高于对照组和脓毒血症组(P<0.05);脓毒血症组与对照组比较,差异无统计学意义(P>0.05);Oglu组β-catenin、TCF-4及MMP-13表达水平较脓毒血症组降低(P<0.05)。结论 Oglu对脓毒血症大鼠空肠损伤发挥保护作用,可能与HIF-1α、Wnt/β-catenin信号通路调控,减轻脓毒血症期间氧化应激及炎症反应有关。 Objective To investigate whether oatβ-glucan(Oglu)can provide protection for jejunum injury with sepsis.Methods Fifty SD rats were randomly divided into a normal control group(n=10),a sepsis group(n=20)and an Oglu group(n=20).Septic models in the sepsis group and the Oglu group were induced with cecal ligation and puncture(CLP).The jejunum injury was assessed by pathology.And the inflammatory cytokines IL-6,TNF-αand procalcitonin were detected by ELISA.The expressions of hypoxia-inducible factor-1(HIF-1α),β-catenin,T-cell factor(TCF-4)and matrix metalloproteinase 13(MMP-13)were detected by PCR and Western blot.Results Oglu relieved sepsis-induced jejunum injury,and reduced the levels of IL-6,TNF-αand procalcitonin in the sepsis group(P<0.05).The expression of HIF-1αin the jejunum of the Oglu group was higher than that of the normal control group and the sepsis group(P<0.05),but there was no significant difference between the latter two groups(P>0.05).The expressions ofβ-catenin,TCF-4 and MMP-13 in the Oglu group was significantly lower than that in the sepsis group(P<0.05).Conclusions Oatβ-glucan exerts protective effect on jejunum in septic rats,which is related to the regulation of HIF-1αand Wnt/β-catenin signaling pathway and the reduction of oxidative stress and inflammatory response during sepsis.
