摘要
目的探讨miR-181d对LCRG1的调控作用及miR-181d对喉癌Hep2细胞增殖的影响。方法利用基因芯片和RTPCR技术检测miR-181d在喉癌组织和癌旁组织中的表达;生物信息学预测喉癌候选抑瘤基因LCRG1的靶标miRNAs;构建LCRG1 3'UTR荧光素酶载体,双荧光素酶检测系统测定其荧光素酶活性;将miR-181d mimic和miR-181d inhibitor瞬转入Hep2细胞,RT-PCR结果验证miR-181d的表达后,再用Western blot法检测各组中LCRG1蛋白的表达;采用MTT实验、迁移、侵袭及流式细胞术等实验,观察转染组与对照组细胞的增殖。结果 miR-181d在喉癌组织中的表达量较癌旁组织明显升高(P=0. 046 5);miR-181d可能靶向结合LCRG1,且LCRG1蛋白表达与miR-181d表达呈负相关;下调miR-181d能降低Hep2细胞的增殖、迁移、侵袭能力,并使细胞周期主要阻滞于G1期。结论在Hep2细胞中miR-181d可以结合LCRG1 3'UTR,负性调控LCRG1的表达并使Hep2细胞增殖、迁移和侵袭能力增强。
Purpose To study the effect of miR-181d on the regulation of tumor suppressor gene LCRG1 and the proliferation of Hep2 cells in laryngeal carcinoma.Methods The expression of miR-181d was detected in laryngeal carcinoma tissues and adjacent normal tissues by miRNAs microarray and real-time PCR(RT-PCR).Bioinformatics predicted that miR-181d might be involved in LCRG1 gene expression.Furthermore,LCRG1 3′UTR luciferase vector was constructed and the luciferase activity was measured by dual luciferase assay system.miR-181d mimics and miR-181d inhibitor was used to transfect Hep2 cells instantaneously,RT-PCR to assess the miRNA expression in Hep2 cells,Western blot assay to validate the protein level of LCRG1,MTT assay,wound scratch assay,migration and invasion experiments to investigate the ability of cell proliferation and apoptosis between transfection group and control group.Results The expression of miR-181d in laryngeal squamous cell carcinoma was significantly higher than that in para-cancerous tissue(P=0.046 5).LCRG1 was directly targeted by miR-181d,the proliferation,migration and invasion of Hep2 cells transfected with miR-181d inhibitor were reduced significantly,and the expression of LCRG1 protein was markedly increased.Conclusion miR-181d can bind to LCRG1 3′UTR in Hep2 cells,which can negatively regulate the expression of LCRG1 and enhance the proliferation,migration and invasion of Hep2 cells.
