摘要
目的分析烧伤复合海水浸泡大鼠的多器官损伤情况。方法将120只大鼠随机分为正常对照组、单纯烧伤组及烧伤浸泡2、6、12、24 h组(n=20)。正常对照组不作处理;单纯烧伤组、烧伤浸泡组制备烧伤模型,烧伤浸泡组烧伤后分别于人工海水中浸泡2、6、12、24 h。将大鼠麻醉后取静脉血,检测肝功能指标、肾功能指标、组织代谢及缺氧指标、血离子。将大鼠处死后取肝、肺、肠组织,HE染色法观察肝、肺、肠组织病理学变化,免疫组化法检测肝、肺、肠组织中凋亡相关蛋白Caspase-3、Bcl-2表达。结果烧伤浸泡组肝功能、肾功能、氧化应激、血离子等部分指标检测结果与正常对照组、单纯烧伤组比较差异有统计学意义(P均<0. 05)。烧伤浸泡组肝、肺及肠组织结构发生损伤。与正常对照组、单纯烧伤组比较,烧伤浸泡组肝、肺及肠组织相关凋亡标志物Casepase-3蛋白阳性表达率高(P均<0. 05)、Bcl-2蛋白阳性表达率低(P均<0. 05)。结论大鼠烧伤后经海水浸泡伤情加重,其肺、肝、肾、肠等主要脏器均发生损伤。
Objective To observe the multi-organ injury of rats with burns and seawater immersion.Methods Totally 120 rats were randomly divided into the normal control group,simple burn group,and burn and seawater immersion for 2,6,12,and 24 h groups(n=20).The rats in the normal control group were not treated;burn model was prepared in the simple burn group and the burn and seawater immersion groups.Following burns,the rats in burn and seawater immersion groups were immersed in artificial seawater for 2,6,12,and 24 h respectively.The rats were anesthetized,then venous blood samples were taken to analyse liver function,renal function,tissue metabolism,hypoxia,and blood ion.The liver,lung and intestine tissues were taken after the rats were sacrificed.The pathological changes of liver,lung and intestine were observed with HE staining,and immunohistochemistry was used to detect the expressions of apoptosis-related proteins Caspase-3 and Bcl-2 in livers,lungs and intestines.Results Some parameters of liver,kidney,oxidative stress,blood ion and other tests in the burn and seawater immersion groups were significantly different from those in the normal control group or in the simple burn group(P<0.05).Liver,lung and intestinal tissue damages occurred in the burn and seawater immersion groups.Compared with the normal control group and the simple burn group,the expression of Caspase-3 in the liver,lung and intestine tissues of the burn and seawater immersion groups was higher,while the expression of Bcl-2 was lower.Conclusion The burns and seawater immersion in rats may cause severe damage,which may lead to multi-organ injuries.
