甲状旁腺激素PTH(1-34)对骨转移瘤生长的影响

Parathyroid hormone PTH(1-34) on the growth of bone metastases

目的:探讨甲状旁腺激素PTH(1-34)对骨转移瘤生长的影响。方法:选取36只4~5周龄BALB/C小鼠,随机分为正常对照组(12只)、模型组(12只)和实验组(12只),构建小鼠乳腺癌模型,分别给予生理盐水腹腔注射与PTH(1-34)腹腔注射。结果:实验组与模型组小鼠骨转移破坏率差异有统计学意义(P<0.05);实验组小鼠BMD及股骨灰干重比显著低于正常对照组与模型组(P<0.05);实验组P、BSP、β-CTX等血清生化标志物与正常对照组、模型组差异有统计学意义(P<0.05);实验组小鼠肿瘤质量及体积均高于正常对照组(P<0.05)。结论:血清生化指标能够辅助诊断骨转移瘤,PTH(1-34)能够增加骨转移率,加快肿瘤的生长,可为临床PTH(1-34)抗体的研发提供参考。

Objective:To investigate the effect of parathyroid hormone PTH(1-34)on the growth of bone metastases.Methods:40 BALB/C mice of 4~5 weeks age were selected,they were randomly divided into three groups,the frist groups with 14 mices as the normal control group,both the second and thrid group were mices.The mice breast cancer model was constructed by intraperitoneal injection of saline(the model group)and PTH(1-34)intraperitoneal injection(the experimental group).Results:The difference of bone metastasis rate between the experimental group and the model group was statistically significant(P<0.05).The BMD and the dry weight ratio of the femur ash in the experimental group were significantly lower than that in the normal control group and the model group(P<0.05);the serum biochemical markers such as P,BSP andβ-CTX in the experimental group were statistically significant(P<0.05)compared with the normal group and the model group(P<0.05).The tumor mass and volume of mice in the group were higher than those in the control group(P<0.05).Conclusion:Serum biochemical indexes can assist the diagnosis of bone metastases.PTH(1-34)can increase the rate of bone metastasis and accelerate the growth of the tumor.It can provide reference for the research and development of clinical PTH(1-34)antibody.
