摘要
为确定FGF2调节奶牛乳腺分支主要信号通路,首先将青春期奶牛乳腺类器官镶嵌在鼠尾Ⅰ型胶原中并添加FGF2作三维培养;通过FGFR1受体抑制剂(FGF2+PD173074组)、PI3K/AKT信号通路抑制剂(FGF2+LY-294002组)或MAPK/ERK信号通路抑制剂(FGF2+U0126组)阻断信号通路,确定不同处理下84 h时奶牛乳腺分支率;利用Western blot检测处理时间0、15、60、240和600 min时各组p-AKT、AKT、p-ERK和ERK相对蛋白表达量。研究发现,FGF2主要通过MAPK信号通路影响奶牛乳腺分支。文章揭示FGF2在影响奶牛乳腺分支形态发生中起主要作用的下游信号通路,对奶牛乳腺分支形态发生分子机制研究具有推动作用。
To determine the main signaling,mammary epithelial organoids from pubertal dairy cows were embedded in collagen type I.FGF2 was added to the medium.At the same time,inhibitor for FGFR1 receptor(FGF2+PD173074 group),for PI3K/AKT signaling(FGF2+LY-294002 group),or for MAPK/ERK signaling(FGF2+U0126 group)was added to the medium,respectively.The branching rates were determined in three-dimensional culture at 84 h.The relative protein expression levels of p-AKT,AKT,p-ERK and ERK were measured in each group at 0,15,60,240,and 600 min by Western blot.It showed that FGF2 affected dairy cow mammary branching morphogenesis mainly through MAPK signaling.Reveal the main downstream signaling that affected dairy cow mammary branching morphogenesis.This could promote the study of the molecular mechanism of dairy cow mammary branching morphogenesis.
