矮坨坨内生真菌Fusarium oxysporum代谢产物及其抗肝癌活性的研究 被引量:4

摘要 从矮坨坨根部分离得到的一株内生菌,经形态与分子生物学18SrDNA序列分析鉴定为尖孢镰刀菌(Fusarium oxysporum).运用中压柱色谱,高效制备液相色谱方法和现代波谱技术1 HNMR、13 CNMR以及LC-MS从其发酵液的乙酸乙酯部位分离鉴定了9个单体化合物,分别为4-氨基-2-羟基嘧啶(1),4,5-二氢-3H-吡唑-3-羧酸甲酯(2),4-羟基-7-(2-羟基-丁基)-3-甲基-氧杂环庚烷-2-酮(3),2-氨基-3-甲基戊酸(4),2,5-二氨基戊酸(5),2-氨基-3-甲基丁酸(6)4-羟基苯甲酸(7),脱氢镰刀菌酸(8),镰刀菌酸(9),其中化合物8对肝癌HepG2和Hep3B细胞具有较强的细胞毒活性,IC50值分别为23.9和29.1μg/mL,而对正常肝细胞L-02未显示细胞毒性. In this study,the endophytic fungus isolated from the roots of Munronia delavayi Franch.was determined as Fusarium oxysporum based on fungus s morphology and 18S rDNA sequence analysis.The isolation and purification of the ethyl acetate extract of fermentation was performed with medium-pressure column chromatography and preparative HPLC.The structures of the nine compounds were identified by extensive spectroscopic analysis as 1HNMR,13 CNMR and LC-MS.Their structures were elucidated as 4-amino-2-hydroxypyrimidinel(1),4,5-Dihydro-3H-pyrazole-3-carboxylic acid methyl(2),4-hydroxy-7-(2-hydroxy-butyl)-3-methyl-oxepan-2-ketone(3),2-amino-3-methyl-pentanoic acid(4),2,5-diamino-pentanoic acid(5),2-amino-3-methyl-butyric acid(6),4-hydroxy-benzoic acid(7),dehydrofusaric acid(8)and fusaric acid(9).Compound 8 exhibited cytotoxic activity against HepG2 and Hep3B cell lines with IC 50 values of 23.9 and 29.1μg/mL,respectively.However,the cytotoxicity was not shown in L-02 of normal liver cells.
